Multilocus sequence typing of Salmonella strains isolated from food poisoning in Shijiazhuang during 2011-2016

摘要 目的探讨多位点序列分型(MLST)方法在沙门菌分型鉴定研究中的应用,了解石家庄地区2011年-2016年食物中毒沙门菌株的分子流行病学特征。方法对32株食物中毒沙门菌进行血清分型复核,并选取沙门菌的7对管家基因aro C、dna N、hem D、his D、pur E、suc A和thr A作为靶标基因进行PCR扩增、测序及序列分析。结果 32株沙门菌分为7个血清型和6个ST型,优势型别为肠炎沙门菌和ST11,发现2个新ST型。沙门菌血清型与MLST型别关系密切,但两者并非完全对应。结论 MLST分型和血清学分型配合使用,有助于深化研究沙门菌的进化关系和溯源调查。 Objective To investigate the application of multilocus sequence typing(MLST) in the classification and identification of Salmonella,and to understand the molecular epidemiological characteristics of Salmonella strains isolated from food poisoning in Shijiazhuang during 2011-2016. Methods Serotyping of 32 Salmonella strains from food poisonings was performed. Seven house-keeping genes of Salmonella(aro C,dna N,hem D,his D,pur E,suc A and thr A) were selected as target genes for PCR amplification,sequencing and sequence analysis. Results Thirty-two isolates were divided to 7 serotypes and 6 STs. The dominant types were Salmonella enteritidis and ST11,and two new ST types were found. Salmonella serotype was closely related to the MLST type,but both of them didn’t correspond exactly. Conclusion The combined use of MLST typing and serological typing will be helpful to further studying the evolutionary relationship and traceability of Salmonella.
