摘要
MicroRNA(miRNA)是在真核生物中发现的一类内源性的具有调控功能的非编码小RNA,在鸡的许多生物学功能中发挥重要作用,但在鸡的应激性免疫抑制的分子调控机制中尚不完全清楚。本试验通过在日粮中添加外源皮质酮激素,构建雏鸡应激性免疫抑制模型,高通量Solexa测序技术建立雏鸡法氏囊差异miRNA表达谱,共鉴定出77个差异表达的miRNAs,34个上调,43个下调,生物学分析预测差异表达miRNA的靶基因有282个,对这些预测的靶基因进行GO和KEGG通路富集分析,结果显示这些通路主要富集到心肌收缩、精氨酸和脯氨酸代谢、RNA聚合酶、肾素-血管紧张素系统。试验结果为揭示家禽应激性免疫抑制转录后分子调控机制提供了依据。
MicroRNA (miRNA) is an endogenous regulatory non-coding small RNAs which is found in eukaryotes and plays an important role in many biological functions of chickens. However, the molecular mechanism of its function in immunosuppression under stress in chick- ens is still unclear. In order to study the Corticosterone-induced miRNAs expression in bursa of Fabricius, corticosterone was added to the basic ration of Gushi chickens to construct a stress immunosuppressive model of chickens. The Solexa sequencing technique was used to es- tablish a differential miRNA expression profile of chicken bursa. 77 differentially expressed miRNAs were identified, of which 34 were up- regulated and 43 were down-regulated. 282 target genes were predicted by a hioinformatics software, which was an analysis by GO ( Gene Ontology) and KEGG pathway enrichment. The results suggested that these pathways were mainly involved in myocardial contraction, arginine and proline metabolism, RNA polymerase, renin-angiotensin system and so on. The present study provides a basis for revealing the post-transcriptional molecular regulation mechanism of stress immunosuppression in poultry.
