壳聚糖复合KGF-2突变体温敏敷料研制及初步评价 被引量:5

Preparation and preliminary evaluation of chitosan composite KGF-2 mutant thermal sensitive dressing

摘要 目的研制壳聚糖复合角质细胞生长因子2突变体(keratinocyte growth factor-2 mutant,KGF-2M)温敏敷料,并对其理化特性和药物释放规律进行初步评价。方法利用壳聚糖、壳聚糖季铵盐、β-甘油磷酸钠等主辅料配置不同配方,并与KGF-2M进行复合研制温敏敷料。对凝胶时间、凝胶温度、药物释放速度等指标进行测定,分析该温敏敷料的理化特性。结果通过对壳聚糖和相关辅料配方成分的优化,获得了一种具有温敏特性的壳聚糖-KGF-2M复合敷料,该敷料在环境温度>35℃时,可在10 min内由液态凝胶转为固态凝胶,其中复合的KGF-2M药物4 h溶出>98%,蛋白活性可基本保持稳定。结论该温敏凝胶具有适形性好、保湿、隔离、促进创面愈合的特性,并具有一定的缓释效果,在战时创面修复中具有巨大的应用潜力。 Objective To develop chitosan composite keratinocyte growth factor-2 mutant (KGF-2M) temperature-sen-sitive dressing and evaluate its physicochemical properties and dynamic release rule were used. Methods Chitosan, chitosan quaternary ammonium salt, β-glyeerophosphate and other adjuvant materials to configure different formulations which were compounded with KGF-2M in order to develop temperature-sensitive dressing. Gelling time, temperature, the release rate of KGF-2M and other indicators were measured to analyze the physical and chemical properties of the temperature-sen- sitive dressing. Results Chitosan-KGF-2M composite dressing with temperature-sensitive properties was obtained by optimizing the formulation components of chitosan and related adjuvant materials. When the liquid dressing was above 35℃, it could be converted from liquid to solid gelatin within 10 minutes. The compound KGF-2M released from the gel was more than 98% at 4 h, and its bioactivity remained stable. Conclusion The thermo-sensitive gel has the characteristics of good conformability, moisturizing( moisture), isolation, wound healing, and a controlled release effect, which has great potential in wartime for wound repair.
