miR-130b靶向STAT3参与家兔非特异性消化道紊乱型肠炎的调节 被引量:1

miR-130b Modulates Rabbit Nonspecific Gastrointestinal Disorder Enteritis via Targeting STAT3

摘要 【目的】肠炎是家兔最常见且死亡率最高的消化道代谢疾病,给家兔养殖业带来巨大的经济损失。圆小囊作为家兔特有的免疫器官,在家兔肠炎中起重要的调节作用。本试验通过研究家兔圆小囊中microRNAs(miRNAs)的表达差异,探究miRNAs与家兔肠炎发生和调节的分子机制。【方法】本试验以60只新西兰兔为试验样本,采用低纤维饲料(纤维素CF 9%、木质素ADF 13%)诱导家兔非特异性消化道紊乱型肠炎发生,通过临床症状和切片观察将试验样本分为4组(n=9),即健康对照组、诱导健康组、轻微组、严重组;检测家兔圆小囊中miR-130b以及相关炎症因子白介素-22(IL-22)、肿瘤坏死因子(TNF-α)和白介素-18(IL-18)的表达,通过miRBase、targetscan等预测miR-130b与STAT3的靶向关系,同时利用q PCR检测STAT3的表达量。【结果】圆小囊中,IL-22、TNF-α、IL-18表达量上调,严重组显著高于其他组(P<0.05),miR-130b表达随着炎症程度的加深表达显著下调,2个患病组极显著低于2个健康组(P<0.01);STAT3随着炎症程度加深表达量显著上调,2个患病组极显著高于2个健康组(P<0.01);在4个分组中,miR-130b和STAT3呈显著的负相关(r=0.635,P<0.01)。【结论】在家兔非特异性消化道紊乱型肠炎发生过程中,圆小囊中的miR-130b通过靶向STAT3参与炎症的发生并发挥作用。 [Objective] Rabbit enteritis is the most common and the highest mortality of digestive tract metabolic diseases which gives rise to huge economic loss to the rabbitry. As a special immune organ in the rabbit, sacculus rounds plays an important role in the regulation of rabbit enteritis. In this study, the molecular mechanism of occurrence and regulation of rabbit enteritis by identifying the expression of microRNA (miRNA) in rabbit sacculus rounds was investigated. [Method] In this experiment, 60 New Zealand rabbits were treated with low dietary fibre (cellulose and lignin CF 9% ADF 13% ) to induce Nonspecific gastrointestinal disorder enteritis, and the rabbits were divided into 4 groups based on clinical symptoms and patbologica, namely health control, health, mild, severe; Then, the expression of miR-130b, inflammatory cytokines interleukin-22 (IL-22), tumor necrosis factor (TNF-α) and interleukin-18 (IL-18) in sacculus rounds were detected, and the relationship between miR-130b and STAI3 was identified with miRBase and targetscan, simultaneously, the expression of STAI3 by qPCR was detected. [Result] The levels of IL-22, TNF-, IL-18 and expression were up-regulated in severe group and higher than others (P〈0.05) and the expression of miR-130b in the sacculus rounds was significantly decreased along with the level of inflammation, and 2 disease groups were significantly lower than the 2 health groups (P〈0.01). Along with the level of inflammation STAI3 expression was upregulated, 2 disease groups were significantly higher than the 2 healthy groups (P〈0.01). There was a significantly negative correlation between miR-130b and STAI3 in the 4 groups (r-0.635, P〈0.01 ). [Conclusion] In the process of rabbit nonspecific gastrointestinal disorder enteritis, miR-130b in sacculus participates in the occurrence of inflammation by targeting STAI3.
