结核分枝杆菌Rv3607c B细胞表位预测及分析

摘要 预测和筛选结核分枝杆菌Rv3607c的B细胞表位,预测和分析得出其优势B细胞线性表位和构象性表位。在NCBI数据库中获得结核分枝杆菌的Rv3607c蛋白序列,使用生物信息学在线软件辅助分析。采用SSPro方案预测Rv3607c蛋白二级结构,Bcepred软件分析Rv3607c各个氨基酸的亲水性、柔韧性、可及性、转角、极性、抗原性等物理化学性质预测蛋白B细胞线性表位,ABCpred服务器利用人工神经网络预测抗原序列中的线性B细胞抗原决定区域,综合SSPro、Bcepred和ABCpred的分析情况进行预测,确定了3个优势B细胞线性表位,分别为:GAEIADHVMDDQRVHA77-92、AAEIVAGPPRKLIETV61-76、GRHGVYDHERVAGQRF14-29。另使用EpiSearch方案利用噬菌体展示肽序列自动检测得到优势构象性表位1个,即G9,T11,G14,G26,R28,V30,D32,A62,I64,A66,I73,E74,T75,A96,D110,V111,A112,V113。笔者对Rv3607c蛋白的B细胞线性表位及构象性表位进行分析并获得多个可能的优势B细胞表位,以期为结核病预防性疫苗的制备及诊断靶标筛选提供参考。 The B-cell epitope of Rv3607 cwas predicted and screened,and its dominant B cell linear epitope and conformational epitope were predicted and analyzed.The Rv3607 cprotein sequence of Mycobacterium tuberculosis was obtained in NCBI database,and the bioinformatics online software was used to assist analysis.SSPro was used in predicting protein secondary structure of Rv3607 c.To predict protein B cell linear epitope,Bcepred software analyses Rv3607 c's each amino acids of physical and chemical properties,such as hydrophilic,flexibility,accessibility,corner,polarity,antigenicity.ABCpred server uses artificial neural network to predict antigen in a sequence of linear B cell regional antigen.According to SSPro,Bcepred and ABCpred projections on analysis,there are three advantaged B cell linear epitope,which are GAEIADHVMDDQRVHA77-92,AAEIVAGPPRKLIETV61-76,GRHGVYDHERVAGQRF14-29.In addition,with thehelp of EpiSearch scheme using phage display peptide sequence automatically,we detected advantaged conformational epitope,it is G9,T11,G14,G26,R28,V30,D32,A62,I64,A66,I73,E74,T75,A96,D110,V111,A112 and V113.This study about protein B cell linear epitope and conformational epitope of Rv3607 cwere analyzed and gained multiple possible advantaged B cell epitope,make preparation of prophylactic vaccine for tuberculosis(TB)and provided the reference for diagnostic target selection.
