降低软枣猕猴桃初代培养污染率的研究

摘要 [目的]筛选降低软枣猕猴桃外植体初代培养污染率和提升外植体诱导率的处理方法。[方法]采用半木质化的软枣猕猴桃新梢作为外植体,根据外植体灭菌时间和氨苄青霉素培养基浓度差异组合,分析降低软枣猕猴桃外植体初代培养污染率和提升外植体诱导率的因素,筛选出最适宜的处理方法。[结果]增加外植体75%乙醇灭菌时间至10 s可降低软枣猕猴桃初代培养污染率,培养基中氨苄青霉素浓度为100~150 mg/L时,软枣猕猴桃初代培养污染率随培养基中氨苄青霉素浓度的增加而降低。增加外植体灭菌时间和培养基中氨苄青霉素浓度均能增加软枣猕猴桃外植体诱导率。[结论]污染率低且诱导率高的处理方法为1/2MS+100 mg/L氨苄青霉素、1/2MS+150 mg/L氨苄青霉素、外植体75%乙醇灭菌10 s+0.1%升汞灭菌8 min和外植体75%乙醇灭菌10 s+0.1%升汞灭菌10 min。 [Objective]To screen the most suitable treatment for reducing Actinidia arguta primary culture contamination rate and increasing explant induction rate.[Method]Semi-woody shoots were used as explants to screen the most suitable treatment for reducing A. arguta primary culture contamination rate and increasing explant induction rate by combining sterilization time and ampicillin medium concentration. [Result]Increasing the sterilization time to 10 s could reduce the contamination rate of A. arguta primary culture,and the contamination rate of primary culture decreased with the increasing of ampicillin concentration during 100 ~ 150 mg/L. Increasing explant sterilization time and ampicillin concentration in culture medium could increase the induction rate of A. arguta.[Conclusion]1/2 MS + 100 mg/L ampicillin,1/2 MS + 150 mg/L ampicillin,explants were sterilized with 75% alcohol with 10 s + 0. 1% mercuric chloride for 8 min and explants 75% alcohol sterilization 10 s +0. 1% mercury chloride sterilization 10 min are the suitable methods with low contamination rate and high induction rate.
