一株野生大型真菌的菌种分离及ITS序列鉴定 被引量:12

摘要 [目的]寻找适宜杨树林下栽培的野生食用菌菌种。[方法]对采集自辽宁省阜新市蒙古族自治县银中杨林下的野生大型真菌进行菌种分离,同时研究不同分离部位对菌丝生长的影响,并对获得菌丝进行了rDNA ITS序列分析。[结果]菌盖与菌柄连接处为菌种最佳分离部位。对分离株rDNA ITS片段进行PCR扩增,扩增产物长度为692 bp,登录NCBI与GenBank中已知的菌种进行Blast比对,序列与Agaricus bitorquis(Quél.)Sacc的Query cover为97%~100%,ident均为99%。[结论]鉴定分离菌种为蘑菇科、蘑菇属、大肥菇。该研究可为下一步开展杨-食用菌复合经营提供菌种准备。 [Objective]To find suitable strains of wild edible fungi cultivated under poplar forests. [Method]Wild macrofungus under Populus alba forest collected from Mongolia Autonomous County,Fuxin City,Liaoning Province was isolated and the effects of different separation sites on mycelium growth were studied. The rDNA ITS sequences of mycelium were studied. [Result]The junction between the cap and stipe was the best separation site for the fungus. The rDNA ITS fragment was amplified by PCR,the length of the amplified product was 692 bp. The Blast comparison was performed between NCBI and a known strain in Gen Bank. The Query cover between sequence and Agaricus bitorquis( Quél.) Sacc was 97% to 100%,the idents were all 99%.[Conclusion]The identified strains belong to mushroom family,mushroom genus and Agaricus bitorquis. The study can provide the preparation of strains for the nest stage of poplar-edible fungus compound management.
