摘要
目的探讨NF-κB/P65信号通路在子宫内膜癌中与自噬、凋亡以及ER、PR之间的关系。方法采用免疫组化和Western blot法检测53例子宫内膜癌与癌旁组织中的NF-κB/P65、自噬相关蛋白Beclin-1、LC3及凋亡相关蛋白Caspase-8、Survivin的表达,并与临床资料以及ER、PR的表达进行了统计学分析。结果与癌旁组织相比,Beclin-1、LC3、Caspase-8在子宫内膜癌组织中表达明显减少,差异具有统计学意义(P<0.05,P<0.01,P<0.05);而癌组织中,NF-κB/P65和Survivin虽与癌旁差异无统计学意义,但已见明显的升高趋势。且结合临床资料分析发现,Beclin-1、LC3的表达均与ER相关(P<0.05),Survivin、Caspase-8的表达与ER、PR相关(均为P<0.05)。结论 NF-κB/P65通过抑制自噬与凋亡,促进子宫内膜癌的发生与发展;ER、PR表达结合NF-κB/P65及自噬与凋亡指标的表达,对雌激素依赖性子宫内膜癌的临床治疗及预后判定具有重要意义。
Objective To investigate the relationship between NF-κB/P65 signaling pathway,autophagy,apoptosis,ER and PR in endometrial carcinoma. Methods Immunohistochemistry and Western blot were used to detect the expression of NF-κB/P65,autophagy-related proteins"Beclin-1 and LC3",apoptosis-related proteins"Caspase-8 and survivin"in 53 cases of endometrial carcinoma and adjacent tissues. Then the relationship between them and the clinical data were statistically analysed,especially the expression of ER and PR. Results The expression of Beclin-1,LC3 and Caspase-8 in endometrial carcinoma was significantly lower than that in adjacent tissues(P〈0. 05,P〈0. 01,P〈0. 05,respectively). Meanwhile,although the difference between NF-κB/P65 and Survivin was not statistically significant,but performed a clear upward trend(P〈0. 05). Besides,the expression of Survivin and Caspase-8 was respectively correlated with ER and PR(P〈0. 05,P〈0. 05). The expression of Beclin-1 and LC3 was respectively correlated with ER(P〈0. 05). Conclusion NF-κB/P65 inhibiting autophagy and apoptosis,promotes the occurrence and development of endometrial cardcinoma; ER and PR expression with the expression of NF-κB/P65,autophagy and apoptosis indexes,has important significance for clinical treatment and prognosis of estrogen dependent endometrial carcinoma.
