摘要
目的研究决明子提取物对高脂血症大鼠血脂的影响。方法选取48只SPF级成年雄性Sprague-Dawley(SD)大鼠,采用饲喂高脂饲料的方法建立高脂血症模型。根据体重随机分为2组,即空白对照组(8只)和高脂血症模型组(40只),给予相应饲料饲养4周后,通过全自动分析生化仪检测动物血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-c)和低密度脂蛋白(LDL-c)的水平。根据TC、TG水平,将高脂血症模型组随机分为5组,即高脂血症模型组,阳性药对照组(阿托伐他汀钙,10mg·kg^(-1)·d^(-1)),决明子提取物低剂量组(54 mg·kg^(-1)·d^(-1))、决明子提取物中剂量组(162 mg·kg^(-1)·d^(-1))、决明子提取物高剂量组(486 mg·kg^(-1)·d^(-1))。各组按10 m L·kg^(-1)灌胃容积给予相应药物,连续灌胃4周。通过HE染色肝脏病理改变。结果高脂饲料饲养4周后,与正常对照组比较,高脂血症模型组大鼠血清TC、TG、LDL-c含量显著升高(P<0.05),证明高脂血症动物模型建立成功。给药4周后,与高脂血症模型组比较,阿托伐他汀钙组、决明子提取物高、中、低剂量组能显著降低大鼠血清中TC、TG、LDL-c的含量(P<0.05)。肝脏组织形态学的结果显示,高脂血症模型组肝细胞胞质内充满大小不等的脂肪油滴,肝细胞内大脂滴将细胞核挤向一侧,少数肝细胞水样变性。与模型组比较,决明子提取物中、高剂量组肝细胞形态较好,细胞质内的脂肪滴数量明显变少。结论决明子提取物可有效调节高脂血症模型大鼠的血脂水平。
Objective To study the effects of Cassiae Semen extract(CSE) to serum lipid level of hyperlipidemia rats. Methods Forty-eight male SPF grade Sprague-Dawley(SD) rats were selected to set up hyperlipidemia model through feeding with high calorie-diet. Rats were randomly divided into 2 groups,normal control group(8 rats) and hyperlipidemia model group(40 rats),according to their body weights. The blood serum levels of TC,TG,HDL-c and LDL-c were detected through automatic biochemical analyzer,after experiment animals were fed with corresponding diets for 4 weeks. Hyperlipidemia model rats were randomly divided into 5 groups,hyperlipidemia model group,positive control group(atorvastatin calcium,10 mg ·kg^-1·d^-1),CSE low dose group(54 mg·kg^-1·d^-1),CSE medium dose group(162 mg·kg^-1·d^-1) and CSE high dose group(486 mg·kg^-1·d^-1),according to the blood serum levels of TC and TG. The liver pathological change was observed through HE staining. Results Compared with normal control group,blood serum levels of TC,TG and LDL-c of hyperlipidemia model group significantly increased(P〈0. 05) after 4 week calorie-diet feeding,which proved the success of hyperlipidemia rat model. Compared with hyperlipidemia model group,atorvastatin calcium and three concentrations of CSE decreased blood serum level of TC,TG and LDL-c significantly. Liver HE staining showed that in hyperlipidemia rats,there were abundant fat droplets in hepatic cytoplasm,nucleuses were in one side of hepatocytes and hydropic degeneration appeared in a few hepatocytes. Compared with hyperlipidemia model group,hepatocytes showed comparative normal morpha,fat droplet amount in hepatic cytoplasm significantly reduced in medium and high dose CSE groups. Conclusion CSE can effectively regulate blood serum levels of TC,TG and LDL-c of hyperlipidemia rats.
