后负荷增加所致心力衰竭模型小鼠气虚证评价 被引量:6

Evaluation of Qi Deficiency Syndrome in Heart Failure Mice Caused by Elevated Afterload

摘要 目的探讨后负荷增加所致的慢性心力衰竭模型小鼠的证候,进行气虚证客观评价。方法350只4周龄SPF级雄性昆明(KM)小鼠随机分为假手术组(150只)和模型组(200只),各组分为术后2、4、6、8、12周亚组,假手术亚组每组30只,模型组亚组每组40只。采用主动脉弓缩窄术制备后负荷增加小鼠心力衰竭模型。收集两组2、4、6、8、12周小鼠宏观表征、一般状态及心脏超声指标,记录两组术后2、4周旷场自主行进总距离。结果与假手术组同期动物比较,模型组小鼠术后2周表现出不同程度的活跃度降低、精神状态不振;术后4周表现出活跃度降低、精神状态不振、目光无神、自主活动减少、抓取反抗降低;术后4周旷场实验自主行进总距离降低(P<0.05);术后6周左室射血分数、左室短轴缩短率降低(P<0.05,P<0.01)。结论术后4~12周模型组小鼠出现与气虚证候相关的宏观表征、一般状态、心脏超声指标的改变,符合气虚证候要素诊断。 Objective To investigate the syndrome nature and objective evaluation of qi deficiency of chronic heart failure mice caused by elevated afterload. Methods Totally 350 KM mice( male,4 weeks old) were randomly assigned to sham-operated( 1 5 0) group and transverse aortic constriction( TAC,200)group. Both groups were randomly divided into 2,4,6,8 and 1 2 weeks subgroup,3 0 in each sham-operated subgroup, 40 in each TAC subgroup. TAC was performed to induce heart failure. Macroscopic indications, general state of mice, as well as echocardiography were observed at 2, 4, 6, 8, 1 2 weeks after surgery respectively. Meanwhile,the distance-moved in open field test was recorded at 2 and 4 weeks after surgery.Results Compared with the mice in sham-operated group,lassitude and decreased activity to different extent were appeared in TAC group at 2 weeks after surgery. Besides reduced activity and lassitude, reduced spontaneous activity and less resistance to grab were observed in TAC group at 4 weeks after surgery. Meanwhile,moved-distance was decreased(P〈0.05). Ejection fraction and shortening fraction of left ventricle were decreased in T AC group( P〈0.05,P 〈0.01) at 6 weeks after surgery. Conclusion Mice with heart failure met the criteria of qi deficiency syndrome including macroscopic indications,general state and echocardiography of mice from 4 weeks to 1 2 weeks after TAC surgery.
