摘要
目的初步筛选食管癌高发区食管鳞癌患者血清中差异表达的小分子RNAs(miRNAs)。方法选择52例食管鳞癌患者(观察组)、52名健康查体者(对照组),均抽取空腹静脉血4 m L。采用AgilentTM高通量miRNA微阵列芯片检测二者血清中miRNAs。结果与对照组比较,观察组26个miRNAs(hsa-let-7b-5p、hsa-miR-107、hsamiR-1207-5p、hsa-miR-1238-3p、hsa-miR-1246、hsa-miR-1290、hsa-miR-15a-5p、hsa-miR-16-5p、hsa-miR-19b-3p、hsamiR-223-3p、hsa-miR-25-3p、hsa-miR-320c、hsa-miR-3663-3p、hsa-miR-4281、hsa-miR-4455、hsa-miR-4516、hsa-miR-451a、hsa-miR-4532、hsa-miR-4634、hsa-miR-4687-3p、hsa-miR-4763-3p、hsa-miR-486-5p、hsa-miR-574-5p、hsa-miR-6090、hsa-miR-6515-3p、hsa-miR-92a-3p)的荧光信号强度FC绝对值均>2,P均<0.05,FDR均<0.1。结论食管鳞癌患者血清中hsa-let-7b-5p、hsa-miR-107、hsa-miR-1207-5p、hsa-miR-1238-3p、hsa-miR-1246、hsa-miR-1290、hsamiR-15a-5p、hsa-miR-16-5p、hsa-miR-19b-3p、hsa-miR-223-3p、hsa-miR-25-3p、hsa-miR-320c、hsa-miR-3663-3p、hsa-miR-4281、hsa-miR-4455、hsa-miR-4516、hsa-miR-451a、hsa-miR-4532、hsa-miR-4634、hsa-miR-4687-3p、hsa-miR-4763-3p、hsa-miR-486-5p、hsa-miR-574-5p、hsa-miR-6090、hsa-miR-6515-3p、hsa-miR-92a-3p差异表达。26个miRNAs可能与食管鳞癌的发生、发展有关。
Objective To screen out the differentially expressed microRNAs(miRNAs) in the serum of esophageal squamous cell carcinoma(ESCC) patients from a high risk area. Methods The blood samples(4 m L per subject) from52 patients with newly diagnosed ESCC(observation group) and 52 age/sex-matched healthy controls(control group) were obtained from Linzhou,Henan province. The miRNAs were detected by AgilentTMhigh-throughput miRNA microarray. Results Compared with the control group,there were 26 up-regulated miRNAs(hsa-let-7 b-5 p,hsa-miR-107,hsa-miR-1207-5 p,hsa-miR-1238-3 p,hsa-miR-1246,hsa-miR-1290,hsa-miR-15 a-5 p,hsa-miR-16-5 p,hsa-miR-19 b-3 p,hsamiR-223-3 p,hsa-miR-25-3 p,hsa-miR-320 c,hsa-miR-3663-3 p,hsa-miR-4281,hsa-miR-4455,hsa-miR-4516,hsa-miR-451 a,hsa-miR-4532,hsa-miR-4634,hsa-miR-4687-3 p,hsa-miR-4763-3 p,hsa-miR-486-5 p,hsa-miR-574-5 p,hsa-miR-6090,hsa-miR-6515-3 p,and hsa-miR-92 a-3 p) in the observation group(FC absolute value 2,all P〈0. 05,all FDR〈0. 1). Conclusion There are 26 differentially expressed miRNAs in the serum of ESCC patients,which may be associated with the occurrence and development of ESCC.
