摘要
目的:探讨氯喹对乳腺癌细胞MCF7的生长抑制作用及其可能机制。方法:取对数生长期的MCF7细胞,用不同浓度氯喹(0、10、20、30、40μmol/L)作用于MCF7细胞不同时间(0 h、24 h、48 h和72 h),通过MTT法检测细胞增殖情况。使用显微镜观察不同浓度氯喹作用MCF7细胞72 h后细胞形态变化。利用Western blot方法检测不同浓度氯喹作用MCF7细胞24 h后细胞内β-catenin蛋白和Cyclin D1蛋白变化情况。结果:与对照组比较,氯喹在一定浓度范围内对MCF7细胞有明显的抑制作用(P<0.05),细胞增殖活性随着剂量和时间增加而逐渐下降。显微镜下观察到氯喹处理MCF7细胞后,细胞数目减少而细胞碎片增加。Western blot结果显示氯喹处理MCF7细胞后,细胞内β-catenin和Cyclin D1蛋白表达出现下调(P<0.05)。结论:氯喹对MCF7细胞增殖具有抑制作用,其机制可能与下调Wnt/β-catenin信号通路有关。
Objective: To investigate the effect of chlorouine on breast cancer cell line MCF7 and its potential mechanism. Methods: MCF7 cells were treated with chlorouine in various concentrations(0,10,20,30,40 μmol/L) for different durations(0 h,24 h,48 h and 72 h).The cells viability was determined by MTT method.The morphological changes of cells incubated with chlorouine after 72 h were observed under the microscope.The expression of β-catenin protein and Cyclin D1 protein was assayed by western blot after MCF7 cells were treated with chlorouine in various concentrations for 24 h. Results: Compared with control group,MCF7 cells proliferation was inhibited in a time and concentration dependent manner when treated with chlorouine( P 〈0.05).The number of cells decreased while cell fragments increased with chlorouine concentration increased.The expression of β-catenin and Cyclin D1was both significantly down-regulated after treatment of chlorouine( P 〈0.05). Conclusion: Chlorouine could inhibit MCF7 cells proliferation and the mechanism may be involved in Wnt/β-catenin pathway.
