摘要
对从厦门地区2种受感染作物上采集的病原菌株进行鉴定分析,为病害防治提供参考。从厦门市同安国家农业科技园区采集受感染的植物样本,平板分离纯化菌株,提取菌株的总DNA,然后用ITS1和ITS4引物PCR扩增,获得其ITS序列并测序。在Genbank进行比对,采用Paul~*4.0构建进化树。从花生和草莓的地上部分采集到相似菌株,PCR扩增得到的ITS序列相同,Genbank比对与进化分析结果均显示该株菌与已报道的Poitrasia circinans菌株在ITS序列上存在较高相似性。本研究得到的感染花生和草莓的菌株为同一株菌,属于Poitrasia circinans,命名为Poitrasia circinans hxfq.1,这也是福建省首次报道Poitrasia circinans感染作物。
To identify the similar phytopathogens purified from two infected crops in Xiamen area,for plant prevention and control were diseased. The infected plants were collected from Tongan National Agricultural Science and Technology Park,Xiamen. Microorganisms in plants were isolated by plate culture. The total DNA of microorganisms were isolated and purified,and the ITS were sequenced after amplified by PCR with ITS1 and ITS4 primers. The ITS sequences were compared to the others in Genbank,and the phylogenetic tree based on ITS was constructed with Paul* 4. 0. 2 similar phytopathogens were purified from the aboveground parts of peanut and strawberry. The ITS sequences of these 2 strains were identified to be the same. The sequence alignments and phylogenetic analysis revealed a high degree of similarity between Poitrasia circinans strains and the isolated strain in ITS. The phytopathogen which infected both peanut and strawberry is the same strain of Poitrasia circinans,named Poitrasia circinans hxfq. 1,and this is the first report of Poitrasia circinans infected crops in Fujian Province.
