江西原生态种植甘薯根际细菌的溶磷、产生长激素及抑菌能力 被引量:1

Phosphate solubilization capacity,producing IAA and their anti-bacteria activity of rhizosphere bacteria from native sweetpotato in Jiangxi

摘要 为探讨甘薯根际细菌的溶磷、产生长激素(IAA)能力,及筛选对甘薯黑斑病具有抑制活性的菌株,采用5种培养基分离江西原生态种植的甘薯根际细菌,并通过16SrDNA序列分析进行菌株的初步鉴定,测定菌株的溶磷作用,分泌植物生长激素的能力和对甘薯黑斑病菌的抑菌能力.从甘薯根际共分离到可培养细菌菌株653株,鉴定的菌株128株,分属于13目20科36个属,其中,伯克氏菌目(Burkholderiales)的细菌最多,占比43.7%;菌株较多的属为马赛菌属(Massilia)、链霉菌属(Streptomyces)和伯克氏菌属(Burkholderia),分别为28,18和13株.供试的98株甘薯根际细菌中具有溶磷效应的细菌较少,占比8.2%,其中溶磷效果较好的菌株为klbmp 11-11,klbmp 13-6和klbmp 15-37;而能产IAA的细菌较多,占根际细菌的63.3%,IAA分泌量在23.72~130.51μg/mL的菌株有28株,以菌株klbmp 6-34的分泌量最高;通过抑菌试验可发现共有18株菌株对甘薯黑斑病菌有不同程度的抑菌效果,其中以菌株klbmp 4-8和klbmp 14-1的抑菌效果最明显,可作为防治甘薯黑斑病菌的优良菌株. In order to explore the function of phosphate solubilization and IAA production of rhizosphere bacteria of sweetpotato, and inhibitory activity to Cerawcystis fimbriata, 5 kinds of culture medium were used to isolate rhizosphere bacteria ftom native sweetpotato in Jiangxi. Rhizosphere bacteria were identified by 16SrDNA sequence preliminarily, the phosphorus dissolution of the strain was determined by method of phosphate solubilization, Salkowski eolorimetrie assay was used to determine ability of produce IAA and plate eonfrontation method was used to deter mine inhibit effeets to Cerawcystis fimbriata. 653 strains bacteria were isolated ftom the rhizosphere of sweetpota to, and 128 strains were identified, which belong to 13 orders, 20 families and 36 genera. Among them, the baeteria for Burkholderiales were the most and accounting for 43.7%, the genus in large quantity were Massilia, Streptomyces and Burkholderia, the number of bacterial strains were 28, 18 and 13 respectively; Among the 98 test rhizosphere bacteria, the phosphorus solubilizing bacteria were less and accounted for 8.2%. The strains with better phosphorus dissolution were klbmp 11-11, klbmp 13-6 and klbmp 15-37, while 63.3% of rhizosphere bacteria can produce IAA, 28 strains have IAA secretion capacity for 23.72-130.51 μg/mL, and klbmp 6-34 has the highest secretion capacity. A total of 18 strains were found to have different inhibitory effects on Cerawcystis fimbriata, klbmp 4-8 and klbmp 14-1 were the most obvious, and can be used as excellent strains against Ceratocystis fimbriata.
