一种篮式生物反应器内Vero细胞计数方法的建立 被引量:1

Establishment of a method for counting Vero cells in basket bioreactor

摘要 传统的篮式生物反应器因构造特殊,在进行Vero细胞培养时,无法取样进行直接细胞计数。实验通过活细胞计数仪,采用双电极电容法直接测量反应器内的活Vero细胞释放的电容,并将其转换为细胞数量,建立一种间接进行Vero细胞计数的方法。通过反应器内Vero细胞生长繁殖所消耗的葡萄糖量来计算所获得的细胞数,对篮式生物反应器Vero细胞培养阶段进行控制。结果表明,Vero细胞在生物反应器内培养5d,累积消耗葡萄糖230g,所获得的细胞数为1.2×10^(11)~1.3×10^(11)个。建立了Vero细胞数与葡萄糖消耗之间的关系,即每消耗1g葡萄糖可供5.345×10~8个Vero细胞生长。 Traditional basket bioreactor can not be sampled for direct cell count in the culture of Vero cells because of its special structure.A double electrode capacitance method was used to measure the capacitance of living Vero cells in the reactor directly,then the capacitance was converted to the number of cells.An indirect method for Vero cell count was obtained.The number of cells was calculated by the amount of glucose consumed in the growth and propagation of Vero cells in the reactor,to control the culture stage of Vero cell in the basket bioreactor.The result showed that the cells number could reach to 1.2×10^11-1.3×10^11 and consumed 230 g of glucose when the Vero cells were cultured for 5 din the bioreactor.The relationship between Vero cell number and glucose consumption was established,that is,1 g glucose could provide energy for 5.345×10^8 cells growth.
