摘要
建立了黑曲霉(Aspergillus niger)xj菌丝体中糠醛含量测定的条件和方法。菌丝体经乙醚提取,采用Spheriorb C18色谱柱(5μm,4.6 mm×150 mm),流动相为乙睛∶0.1%乙酸水(冰乙酸调)(体积比10∶90),柱温30℃;流速1.0 m L/min,波长λ=277 nm紫外检测器检测的条件下,糠醛呈现良好的分离效果和重现性。其保留时间为5.482 min,最低检出限为0.01 mg/kg,平均回收率为90.51%~93.54%,精密度实验结果相对标准偏差(RSD)为0.98%。测得黑曲霉菌丝体中糠醛含量为1.872 mg/kg,该方法简便、快速、分离效果好,可为今后测定黑曲霉菌丝体中糠醛含量提供一定参考。
The method for the determination of furfural in mycelium of Aspergillus niger xjwas established. The mycelium was extracted by diethyl ether with Spheriorb C18 chromatographic column (4.6 mm×150 mm, 5 μm) as the stationary phase, with acetonitrile : 0.1% acetic acid-water (ice acetic acid)=10:90 (V/V) as mobile phase, column temperature 30 ℃., flow rate 1.0 ml/min, the detection wavelength was 277 nm. Under the chro-matographic conditions, the furfural had good separation effect and reproducibility. The retention time was about 5.482 min, the limit of detection was 0.01 mg/kg, the average recovery rate was 90.51%-93.54%, the relative standard deviation (RSD) of precision test was 0.98%. The content of furfural in mycelium was 1.872 mg/kg. The method was accurate, reliable and reproducible, and could be used as a reference for the determination of furfural content in mycelium of A. niger.
