L-肉碱在罗非鱼体内的药代动力学

摘要 在水温(28±1)℃时,给体质量(82.72±3.65)g和(413.83±39.92)g的吉富罗非鱼口灌剂量12mg/kg的肉碱;另组中给(82.72±3.65)g的吉富罗非鱼口灌剂量12mg/kg和120mg/kg的肉碱,分别于口灌前(0h)和给药后0.083、0.13、0.20、0.25、0.33、0.5、1、2、4、8、12、24、36h和48h采样,数据用WinNonlin 5.2软件非房室模型统计矩方法分析,分别研究了肉碱对不同规格吉富罗非鱼的药代动力学和肉碱剂量对吉富罗非鱼的药代动力学。试验结果表明,口灌给药后,2个剂量组的达峰时间、清除率及平均滞留时间差异不显著(P>0.05),但12mg/kg剂量组的达峰浓度(82.28μmol/L)、药时曲线下面积(431.06h·μmol/L)显著高于120mg/kg剂量组的达峰浓度(21.27μmol/L)、药时曲线下面积(96.95h·μmol/L),说明药物剂量影响肉碱吸收。肉碱在(小规格罗非鱼体内的达峰浓度、清除率和消除半衰期均随个体增大而变小,但差异均不显著(P>0.05)。L-肉碱在大规格罗非鱼体内的分布程度较小规格罗非鱼广泛,即大规格罗非鱼清除率低于小规格罗非鱼,小规格罗非鱼的平均滞留时间比大规格罗非鱼长。 GIFT tilapia(Oreochromis niloticus)with body weight of(82.72±3.65)g(S)and(413.83±39.92)g(L)were administrated orally by L-carnitine at a dose of 12 mg/kg at water temperature of(28±1)℃to evaluate the effects of L-carnitine on pharmacokinetics of GIFT tilapia with different body weight.In other trail,GIFT tilapia with body weight of(82.72±3.65)g(S)were administrated orally by Lcarnitine at a dose of 12 mg/kg and 120 mg/kg to evaluate the effects of L-carnitine dose on pharmacokinetics of GIFT tilapia.The parameters were detected by high performance liquid chromatography(HPLC)and analyzed with the WinNonlin 5.2 software non-compartmental model statistical moment method.The results showed that there were no significant differences in the peak time,clearance rate and mean residence time between the 12 mg/kg and 120 mg/kg(P〉0.05).There were significantly higher area under the curve(431.06 h·μmol/L)and peak concentration(82.28μmol/L)in GIFT tilapia administrated orally by L-carnitine at a dose of 12 mg/kg than those at a dose of 120 mg/kg(21.27 mmol/L,and 96.95 h·μmol/L),indicating that drug dose is involved in L-carnitine absorption.In the same dose of L-carnitine,the peak concentration of L-carnitine in the Tmax,clearance rate(CL)and elimination half-life(t1/2)were decreased with age increase in tilapia,without significant difference(P〉0.05).The large GIFT tilapia had wider distribution level of L-carnitine(area under the concentration time curve)than small tilapia did.
