摘要 对比了重量法与比色法在糠壳果胶质含量测定中的适用性,结果显示比色法更适合糠壳中果胶质的测定。浓硫酸用量12 mL,水解温度85~90℃,水解时间20 min,1.5 g/mL咔唑乙醇溶液1 mL,显色温度25℃左右,显色时间30 min,其相应测定波长530 nm,且不需用乙醇洗涤去糖,在最后测定阶段不需要稀释上清液。本方法简单准确,灵敏度高,重现性好,可作为糠壳果胶质含量的测定方法。 In this study, we compared the applicability of the gravimetric method and the colorimetric method in the determination of pectin content in rice husk. The results showed that the colorimetric method was more suitable for the determination of pectin content in rice husk. The determination conditions were as follows:12 mL concentrated sulfuric acid, hydrolysis at 80 ℃ to 90 ℃ for 20 min,1 mL carbazole-alcohol solution(1.5 g/mL), coloration at 25 ℃ for 30 min, detection wave length of 530 nm. Ethanol washing for sugar removing was not needed, and the supernatant did not need attenuating in the detection process. This method has the advantages of simplicity, high accuracy, high sensitivity and good repeatability, and can be used for the determination of pectin content in rice husk.(Trans. by HUANG Xiaoli)
