柴胡提取物改善慢波睡眠促进动物增高机制的实验研究 被引量:10

摘要 目的 :探讨柴胡提取物调节慢波睡眠促进动物增高的作用机制。方法 :小鼠灌胃柴胡提取物(0.940 mg/kg),与空白对照组比较,观察药物是否促进发育期小鼠体长增长,小鼠脑组织5-羟色胺(5-HT)含量变化;同时给予大鼠药物检测慢波睡眠的变化及体内生长激素含量变化。结果:柴胡提取物可以促进发育期小鼠体长增长,灌胃10 d和20 d后用药组体长变化与空白组比较差异有统计学意义(P〈0.01),脑组织5-HT含量升高与空白组比较差异有统计学意义(P〈0.01)。用药组大鼠慢波睡眠延长,与空白组比较差异有统计学意义(P〈0.05),同时用药组血清生长激素含量升高,与空白组比较差异有统计学意义(P〈0.01),而5-HT选择性抑制剂可以抑制这种趋势。结论:柴胡提取物可以促进动物体长的增长,其作用机制可能是通过提高脑组织5-HT含量升高,延长慢波睡眠时长,从而促进生长激素的分泌引起的。 Objective:To explore the mechanism of Radix bupleuri extract in promoting the growth of animal body length by adjusting slow wave sleep. Methods:The mice in the medication group was given the Radix bupleuri extract(0.940 mg/kg) and compared with the control group. Changes in the growth of the body length and the content of 5-HT in the brain tissues of mice were observed. At the same time,the rats were also given Radix bupleuri extract,and the changes of slow-wave sleep and changes of growth hormone levels were detected. Results:The extract of Radix bupleuri can promote the body length growth of mice at development stage. After 10 days and 20 days of gavage,the change of body length in the medication group was statistically different compared with the blank group(P〈0.01). The increase of 5-HT content in the medication group was statistically different compared with the blank group(P〈0.01). The slow wave sleep prolongation in the treatment group was statistically different compared with that in the blank group(P〈0.05),and the growth hormone level in the medication group increased significantly compared with the blank group(P〈0.01). 5-HT selective inhibitors can inhibit this change. Conclusion:Radix bupleuri extract can promote the growth of animal body length,and this may be due to the promotion of growth hormone secretion caused by increased 5-HT levels in brain tissue and prolonged slow wave sleep duration.
