多枝雾水葛中总黄酮的AB-8大孔树脂纯化工艺研究 被引量:5

摘要 目的:考察大孔树脂富集纯化多枝雾水葛中总黄酮的工艺条件。方法:通过对7种不同型号的大孔树脂进行静态吸附实验,以吸附量作为指标优选合适的树脂,以多枝雾水葛中总黄酮的含量为指标优化纯化的工艺参数。结果:弱极性的AB-8大孔树脂对多枝雾水葛总黄酮具有较好的吸附性能,其最佳工艺条件为供试液质量浓度为0.3g生药·mL^(-1),调节pH=5.0,流速控制为2BV·h^(-1),用5BV供试液上样,以2BV的水除杂,以2BV的60%乙醇进行洗脱。经树脂富集后的总黄酮含量高达68.06%。结论:AB-8对多枝雾水葛总黄酮的分离纯化效果最佳,优化后工艺条件稳定,适用于多枝雾水葛总黄酮的富集。 Objective:To investigate the purification technology of total flavonoids from Pouzolzia zeylanica(L.)Benn.var.microphylla(Wedd.)W.T.Wang by AB-8 macroporous resin.Methods:As per gram of resin adsorption index,seven macroporous resins were chosen with static adsorption experiments.Study on purification parameters with the content of total flavonoids from Pouzolzia zeylanica(L.)Benn.var.microphylla(Wedd.)W.T.Wang.Results:AB-8 macroporous resin was found to have good adsorption and desorption effects.The optimal purification conditions were pH value of 5.0,sample mass concentration of 0.3 g·mL-1,the loaded amount of 5 BV,and loading flow rate of 2 BV·h-1.The sample was eluted by water and 60%ethanol of 2 BV respectively.The concentration of total flavonoids increased to 68.06%.Conclusion:AB-8 is an ideal resin with the best enrichment for separating and purifying the total flavonoids of Pouzolzia zeylanica(L.)Benn.var.microphylla(Wedd.)W.T.Wang.This optimized process is stable and suitable for separation and purification of total flavonoids fromPouzolzia zeylanica(L.)Benn.var.microphylla(Wedd.)W.T.Wang.
