摘要
试验旨在筛选鸡TLR3、TLR4和TLR21基因中的功能性nsSNPs。利用SIFT、PolyPhen-2、PANTHER、PMut和PROVEAN五种软件方法分析引起的氨基酸替换nsSNPs是否可能影响基因的功能。进一步对三个基因氨基酸序列进行翻译后修饰位点及进化位点保守性的预测,使用easymodeller和SWISS-MODEL构建了其野生型及突变型蛋白质的空间结构,并计算突变前后的RMSD。结果表明:TLR3的K240T、T767S、D849N,TLR4的F427V、K714R和TLR21的T455A、Y553H、L602P、W926S可能严重影响蛋白质的结构功能。
In this study, non-synonymous single nucleotide polymorphisms(SNPs) in chicken TLR3, TLR4 and TLR21 genes were isolated from different angles using a variety of bioinformatics techniques. The functional ns SNPs in chicken TLR3, TLR4 and TLR21 genes were screened by SIFT, Poly Phen-2, PANTHER, PMut and PROVEAN in the SNP database and literature. The amino acid sequence of the amino acid sequence was modified and the conserved site was predicted. The spatial structure of the wild type and mutant protein was constructed by using easymodeller and SWISS-MODEL and to calculate the RMSD before and after the mutation, to further verify the impact of ns SNPs on the corresponding protein. The results showed that K240T, T767S, D849N of TLR3, F427V, K714R of TLR4 and T455A, Y553H, L602P, W926S of TLR21 were predicted most damaging.
