禽腺病毒4型人工感染SPF鸡动态病理学观察 被引量:4

摘要 为了深入研究禽腺病毒4型的致病机理,以34日龄SPF鸡人工感染禽腺病毒4型建立了禽腺病毒感染病理模型,并在感染后第1~10天剖杀试验鸡,采集胸腺、心、肝、腺胃、脾、肾,采用常规病理学方法和免疫组织化学方法研究其在感染SPF鸡体内的动态病理变化规律。结果表明,34日龄SPF鸡经口服途径感染禽腺病毒4型,成功建立了人工感染动物模型。感染后第3天鸡出现临床症状,第4~6天为死亡高峰期,第7天后病鸡开始恢复,发病率75%,死亡率67.8%。攻毒后第5天病鸡肝脏病变最重,病变肝细胞内可见嗜碱性、嗜酸性的包涵体,免疫组织化学染色可见攻毒后第5天鸡的肝细胞、脾脏网状细胞和肾小管上皮细胞中出现棕黄色阳性颗粒。 In order to study the pathogenesis of fowl adenovirus type 4, the pathological model of fowl adenovirus infection was established through artificially infected 34-day-age SPF chicken. The dynamic pathobiological changes of thymus, heart, liver, glandular stomach, spleen, kidney were observed with the ordinary pathobiological method and immunohistochemical method day by day until 10 days after infection. The results showed that the pathological model of fowl adenovirus type 4 infection was successfully established after 34-day-age SPF chicken were artificially infected via oral route. The infected chicken developed symptoms at the 3 rd day after infection, died intensively at the 4 th to 6 th days, and began to recovery after the 7 th day. The incidence was 75% and the mortality rate was67.8%. The liver lesion of infected chicken was the most serious at the 5 th day after infection, with eosinophilic and basophilic inclusion bodies in the pathological hepatocytes. Immunohistochemical staining showed that the claybank positive particle representing the virus was appeared in the hepatocytes, splenic reticular cells and renal tubular epithelial cell of the infected chicken at the 5 th day after infection.
