贵州壮族人群25个Y-STR基因座遗传多态性 被引量:3

Genetic polymorphism of 25 Y-STR loci in Zhuang population in Guizhou

摘要 调查贵州黔东南苗族侗族自治州从江县壮族无关男性个体的25个Y-STR基因座的遗传多态性,并探讨与其他12民族群体的遗传关系。采用Y27plex荧光检测试剂盒复合扩增67例样本,采用3130XL遗传分析仪进行毛细管电泳检测,使用Arlequin 3.5、Mega 7.0、SPSS19.0等一系列遗传学软件对基因分型结果进行统计。除2个双拷贝基因座DYS385ab、DYS387S1分别检出8、10个等位基因外,其余23个单拷贝基因座共检出86种等位基因,等位基因个数为2~7不等,等位基因频率为(0.014 9~0.970 1),基因多样性GD值为0.058 8(DYS456)~0.823 6(DYS387S1),25个Y-STR基因座组成的单倍型共检出36种,其中66.7%为唯一单倍型,单倍型多样性HD值为0.957 9。Rst遗传距离矩阵显示:贵州壮族人群父系遗传结构与文献中的福建畲族、内蒙古的蒙古族、山西汉族人群较为相似,与广西壮族、云南哈尼族、新疆维吾尔族、辽宁满族、吉林朝鲜族、萨尔茨堡人群差异明显。而与新疆哈萨克族、安多藏族、西非人群差异显著。25个Y-STR基因座在贵州壮族人群中具有良好的遗传多态性,可应用于父系亲权关系鉴定。 To determine polymorphic features of 25 Y chromosome Short Tandem Repeats (Y-STR ) loci in the Zhuang ethnic and their differences from the corresponding features in other ethnics province, we analyzed the 25 Y-STR loci in the Zhuang ethnic population to obtained of the ethnic group. We amplified 25 Y chromosome Short Tandem Repeats loci from the DNA samples of 67 people of Zhuang ethnic using an Y27 Plex fluorescence detection kit , follow^ed by electrophoresis with a Genetic Analyzer. Genotyping results were analyzed with a series of genetic software , including Arlequin 3. 5 , Mega 7. 0 , and SPSS19. 0. Two double-copy loci DYS385ab and DYS387S1 were found to have 8, 10 alleles, whereas the remaining 23 single-copy loci were found to have 86 different kinds of alleles with the allele number varying from 2 to 7, the allele frequency ranging from 0. 014 9 to0. 970 1, and the genetic diversity varying fom 0. 058 8 (for DYS456) to 0. 823 6 (for DYS387S1) . Atotal of 36 haplotypes were detected, of which 66. 7% were unique. The haplotype diversity was 0. 9579. The Rst genetic distance matrix showed that the genetic structure of the paternal network of the Zhuangethnic group in Guizhou was similar to that of the Fujian She ethnic and Inner Mongolia polymorphism! of the 25 Y-STR loci in Guizhou Zhuang ethnic group differed significantly from that ofthe Yunnan Hani , Xinjiang Uygur , Liaoning Manchu , Jilin Korean and Salzburg , with the greatest difference from that of the Xinjiang Kazak,Amdo Tibetan,and West African people. The 25 Y-STR loci in the Guizhou Zhuang ethnic group had great genetic polymorphisms,which can be used in of parental relationship.
