黄芩对慢性肾衰竭大鼠微炎症状态的影响 被引量:9

Effects of scutellaria on micro-inflammatory state in chronic renal failure rats

摘要 目的研究黄芩对慢性肾衰竭(CRF)大鼠微炎症状态的调节作用。方法将40只Wistar大鼠随机分为4组,即正常组、模型组、条芩组和枯芩组,每组各10只。除正常组外,其余3组大鼠均采用5/6肾切除法制备CRF模型。造模完成后4周,条芩组、枯芩组大鼠分别予相应的药液灌胃,给药量为2.7 g生药(4.5 mL药液)/(kg·d),正常组及模型组分别予等容积0.9%氯化钠注射液灌胃,均每日1次,连续12周。分别在灌胃前(造模完成后4周),灌胃后4、8、12周测量4组大鼠体质量,观察实验过程中4组大鼠精神状态、活动度、反应、体毛色泽、食欲及大小便等一般情况;比较4组大鼠灌胃前,灌胃后4、8、12周血肌酐(Cr)、尿素氮(BUN)、白细胞介素6(IL-6)及C反应蛋白(CRP)变化情况,最后处死各组大鼠,观察肾组织病理变化。结果条芩组和枯芩组大鼠一般状态及肾组织病理改变较模型组轻。灌胃8周后,条芩组Cr、BUN水平及枯芩组Cr、BUN、IL-6、CRP水平均明显低于模型组(P<0.05),且枯芩组IL-6水平低于条芩组及正常组(P<0.05)。灌胃12周后,条芩组Cr、IL-6、CRP水平及枯芩组Cr、BUN、IL-6、CRP水平均明显低于模型组(P<0.05)。结论黄芩能有效降低CRF大鼠血清IL-6和CRP水平,改善微炎症状态可能是其延缓肾衰竭进展的重要作用机制之一。 Objective To investigate the regulatory effect of scutellaria on microinflammatory state in chronic renal failure( CRF) rats. Methods Forty Wistar rats were randomly divided into normal group,model group,radix scutellariae group,and dried scutellaria group. Each group including ten rats. In addition to the normal group,the other three groups of rats were all prepared with a 5/6 nephrectomy to prepare a chronic renal failure model. Four weeks after the completion of the modeling,the rats in radix scutellariae group and dried scutellaria group received 2.7 g crude drug( 4. 5 mL drug solution)/( kg·d) for intragastric administration,and normal group and model group received equal volume 0. 9 % sodium chlorine injection was given to the stomach once daily for 12 consecutive weeks.The body mass of four groups of rats was measured before gavage( 4 weeks after the completion of the model) and at4 weeks,8 weeks and 12 weeks after gavage. The general conditions of mental state,activity,reaction,body hair color,appetite and stool in the 4 groups of rats were observed. The changes of blood creatinine( Cr),urea nitrogen( BUN),leukocyte 6( IL-6) and C reactive protein( CRP) and pathological changes of renal tissue blood creatinine( Cr) before gavage,and 4 weeks,8 weeks and 12 weeks after gavage were observed between four groups. Results The general state and pathological changes of the rats in radix scutellariae group and dried scutellaria group were better than that in the model group. After 8 weeks of gavage,the levels of Cr and BUN in radix scutellariae group and the levels of Cr,BUN,IL-6 and CRP in dried scutellaria group were significantly lower than those in the model group( P〈0. 05),and IL-6 in dried scutellaria group was lower than the radix scutellariae group and normal group( P〈0. 05). After 12 weeks of gavage,the levels of Cr,IL-6,CRP in radix scutellariae group and Cr,BUN,IL-6,and CRP in dried scutellaria group were significantly lower than those in the model group( P〈0. 05). Conclusion Scutellaria can effectively reduce the serum levels of IL-6 and CRP in CRF rats and improve the micro-inflammation state,which may be one of the important mechanisms in delaying the progression of chronic renal failure and reducing complications.
