摘要
目的探讨血脑屏障(BBB)紧密连接相关蛋白claudin 5、occludin及ZO-1在肺癌脑转移瘤中的表达及其临床意义。方法采用免疫组织化学法检测肺癌脑转移瘤组织及正常脑组织中claudin 5、occludin、ZO-1蛋白的表达,并分析紧密连接蛋白表达水平与肺癌脑转移瘤患者临床特征及生存期的关系。结果肺癌脑转移瘤组织中claudin 5、occludin、ZO-1的高表达率均低于正常脑组织(P<0.05);claudin 5、occludin、ZO-1蛋白的表达与肺癌脑转移瘤患者的年龄、病灶个数有关(P<0.05);三种蛋白高表达组患者的中位总生存时间均长于其低表达组患者(P<0.05)。结论紧密连接相关蛋白claudin 5、occludin、ZO-1表达下调可能在脑转移瘤的发生中起到一定的作用。
Objective To investigate the expression of tight junction associated proteins claudin 5, occludin and ZO-1 in brain metastases from lung cancer and their clinical significance. Method Immunohistochemistry assay was used to detect the expression of claudin 5, occludin and ZO-1 protein in brain metastases from lung cancer and normal brain tissues. The correlation between claudin 5, occludin and ZO-1 expression and the clinical characteristics, as well as survival time was analyzed. Result Expression level of tight junction proteins were lower in brain metastasis tissues compared with normal brain tissues(P〈0.05); furthermore, the expression of claudin 5, occludin and ZO-1 showed obvious correlation with patients’ age and the number of lesion(P〈0.05); and high expression level of the three proteins was related to longer median survival time(P〈0.05). Conclusion Decreased expression of claudin 5, occludin and ZO-1 may play an important role in the occurrence of brain metastasis from lung cancer.
