摘要
目的检测miRNA-376c在结直肠癌组织及对应的癌旁正常组织中的表达水平,并分析其与结直肠癌患者临床特征及预后的关系。方法收集202例结直肠癌患者的结直肠癌组织标本及癌旁正常组织标本。按照有无淋巴结转移,将结直肠癌组织分为A组(n=99)和B组(n=103),将癌旁正常组织分为C组(n=99)和D组(n=103);按照TNM分期将结直肠癌患者分为Ⅰ~Ⅱ期(n=101)、Ⅲ期(n=83)和Ⅳ期(n=18)。采用实时荧光定量PCR的方法检测结直肠癌组织和癌旁正常组织中miRNA-376c的表达水平;分析miRNA-376c与结直肠癌患者临床特征及预后的关系。结果 miRNA-376c在结直肠癌组织中的相对表达量为(-10.918±0.101),明显低于癌旁正常组织的(-9.700±0.103)(P<0.01)。A组中miRNA-376c的相对表达量明显高于B组[(-10.620±0.136)vs(-11.230±0.143),P<0.01]。miRNA-376c在Ⅰ~Ⅱ期、Ⅲ期、Ⅳ期患者结直肠癌组织中的相对表达量逐渐升高,分别为(-11.230±0.143)、(-10.790±0.147)、(-9.848±0.300),差异有统计学意义(F=8.22,P<0.01)。高表达的miRNA-376c与结直肠癌患者的不良预后相关。结论高表达水平的miRNA-376c与结直肠癌患者的低分化、高TNM分期、发生淋巴结转移以及不良预后相关,miRNA-376c有可能作为预测结直肠癌患者术后进展及生存期的潜在标志物。
Objective To detect the expression level of miRNA-376c in colorectal cancer tissues and adjacent normal tissues, to analyze its relationship with the clinical characteristics and prognosis of colorectal cancer patients. Method The colorectal cancer tissues and adjacent normal tissues from a total of 202 colorectal cancer patients were collected. According to the presence or absence of lymph node metastasis, colorectal cancer tissues were divided as group A(n=99)and group B(n=103), and adjacent normal tissues were stratified as group C(n=99) and group D(n=103). In respect of TNM staging, the colorectal cancer patients were classified as stage Ⅰ-Ⅱ(n=101), stage Ⅲ(n=83) and stage Ⅳ(n=18).The expression level of miRNA-376c in colorectal cancer tissues and adjacent normal tissues was detected by real-time fluorescence quantitative PCR. The relationship between miRNA-376c and clinical characteristics and prognosis of colorectal cancer patients was analyzed. Result The relative expression level of miRNA-376 c in colorectal cancer tissues was(-10.918±0.101), which was significantly lower than that in adjacent normal tissues(-9.700±0.103)(P〈0.01).The relative expression of miRNA-376c in group A was significantly higher than that in group B [(-10.620±0.136) vs(-11.230±0.143), P〈0.01]. The relative expression levels of miRNA-376 c in stage I-II, III, and IV colorectal cancer tissues were increased gradually, and were(-11.230±0.143),(-10.790±0.147) and(-9.848±0.300), respectively, with statistically significant difference observed(F=8.22, P〈0.01). Highly expressed miRNA-376c was associated with poor prognosis in patients with colorectal cancer. Conclusion High expression levels of miRNA-376c are associated with poor differentiation, high TNM stages, lymph node metastasis, and poor prognosis in patients with colorectal cancer. miRNA-376c may serve as a potential predictor of postoperative progression and survival of patients with colorectal cancer.
