升阳益胃颗粒对慢性疲劳大鼠血清白介素2、γ-干扰素和肿瘤坏死因子α的影响观察 被引量:7

Influence of Shengyangyiwei granule on interleukin-2,interferon and tumor necrosis factor in serum of chronic fatigue rats

摘要 目的探索慢性疲劳综合征(Chronic Fatigue Syndrom,CFS)的中医病机,探讨升阳益胃颗粒发挥抗疲劳作用的机制。方法通过采用慢性束缚、夹尾激怒加强制冷水游泳的复合应激的方法,建立慢性疲劳大鼠模型。造模成功后,每组给予相应药物剂量灌胃,1次/d,连续14d。末次灌胃后测定大鼠血清中白介素-2(IL-2)、γ-干扰素(INF-γ)和肿瘤坏死因子α(TNF-α)的含量。结果模型组大鼠IL-2和INF-γ、TNF-α水平与正常组比较;差异具有统计学意义(P<0.01),而升阳益胃颗粒低、中、高各剂量与模型组比较,IL-2和INF-γ含量均升高,TNF-α含量降低,差异具有统计学意义(P<0.01),且随着剂量浓度的增高,改善各实验指标越明显;与对照组比较,升阳益胃汤颗粒低剂量组对IL-2和IFN-γ的升高作用与对照组作用相当(P>0.05),对TNF-α的降低作用劣于对照组(P<0.05),而中剂量组对IL-2和IFN-γ的升高作用优于对照组(P<0.05),对TNF-α的降低作用与对照组相当(P>0.05),高剂量组对IL-2、IFN-γ的升高作用和对TNF-α的降低作用均明显优于对照组(P<0.01)。结论 (1)升阳益胃颗粒可以明显改善慢性疲劳大鼠的疲劳状态,提高血清白介素2和γ干扰素的含量,降低肿瘤坏死因子α的含量,且随着剂量浓度的增高,其作用越强。 Objective To explore the TCM pathogenesis of chronic fatigue syndrome(CFS). To probe into the mechanism of anti fatigue effect of shengyangyiwei granule. Methods The chronic fatigue rat model was established by using the chronic restraint stress,irritating by tails clamped and forced swimming in cold water. After successful modeling,each group was given intragastric administration of a corresponding dose of medicine,1 times/d,14 consecutive days. The content of interleukin-2(IL-2),interferon-γ(INF-γ) and tumor necrosis factor α(TNF-α) in the serum of rats was measured after the last gastric perfusion. Results The levels of IL-2 and INF-γ and TNF-α in the model group were compared with those in the normal group,and the difference was statistically significant(P〈0. 01). Shengyangyiwei granule low,middle and high dose group compared with model group,IL-2 and INF-γ levels were increased and TNF-α level was decrease,With significant difference(P〈0. 01),and the higher the dose concentration,the more obvious the improvement of the experimental indexes. Compared with the control group,the increase of IL-2 and IFN-γin the low-dose group of shengyang yiwei granule group was comparable to that of the control group(P〉0. 05),and the decrease of TNF-α was inferior to the control group(P〈0. 05). In the medium dose group,the increase of il-2 and IFN-was better than the control group(P〈0. 05),the decrease of TNF-α was comparable to that of the control group(P〉0. 05). High dose group was significantly better than the control group for the increase of il-2 and IFN-and the decrease of TNF-α(P〈0. 01). Conclusion Shengyangyiwei granule can obviously improve the fatigue state of chronic fatigue rats,increase the content of interleukin 2 and interferon-γ and decrease the content of TNF-α in serum,and with the increase of dose concentration,the stronger the effect.
