罗氏内异方对子宫内膜异位症大鼠Bcl-2、Bax表达及内膜组织形态的影响

Effect of Luo 's prescription for endometriosis on Bcl-2 and Bax expressions and morphology of endometriosis

目的探讨罗氏内异方对子宫内膜异位症大鼠Bcl-2、Bax表达及内膜组织形态的影响。方法用自体组织移植法建立子宫内膜异位症大鼠模型,分别以高、低剂量罗氏内异方及达那唑治疗,以对照组及空白组作对照,观察在位内膜、异位内膜的组织形态,并用免疫组化法检测在位内膜、异位内膜Bcl-2、Bax蛋白的表达。结果在位内膜中,低剂量组、达那唑组的Bcl-2蛋白表达明显降低,Bax蛋白表达稍有升高;异位内膜中高剂量组、低剂量组、达那唑组的Bcl-2蛋白表达明显下降,Bax蛋白稍升高。罗氏内异方两组及达那唑可使异位内膜明显变薄、固有层间质萎缩,但达那唑会使在位内膜轻度变性,罗氏内异方则对在位内膜无明显影响。结论罗氏内异方可通过下调内膜细胞Bcl-2的表达,上调Bax的表达,促进内膜细胞的凋亡,从而治疗子宫内膜异位症。

Objective To explore the effect of Luo’s prescription for endometriosis on Bcl-2 and Bax expressions and morphology of endometriosis. Methods Self-endometrium transplantation was used to establish endometriosis rat model,then the rats were treated by high-dose Luo’s prescription for endometriosis,low-dose Luo’s prescription for endometriosis,and danazol,respectively. The rats in control group and blank group were also selected. The morphologies of eutopic endometrium and ectopic endometrium were observed. Immunohistochemical method was used to detect the expressions of Bcl-2 protein and Bax protein in eutopic endometrium and ectopic endometrium. Results In eutopic endometrium,the expression levels of Bcl-2 protein in low-dose group and danazol group decreased significantly,while the expression levels of Bax protein in low-dose group and danazol group increased slightly. In ectopic endometrium,the expression levels of Bcl-2 protein in high-dose group,low-dose group and danazol group decreased significantly,while the expression levels of Bax protein in high-dose group,low-dose group and danazol group increased slightly. In high-dose group,low-dose group and danazol group,the thickness of ectopic endometrium thinned,and mesenchyme in lamina propria atrophied. Danazol induced mild degeneration of eutopic endometrium,while Luo’s prescription for endometriosis had no significant impact on eutopic endometrium. Conclusion Luo’s prescription for endometriosis can treat endometriosis by downregulating Bcl-2 expression,upregulating Bax expression,and promoting cell apoptosis.
