摘要
目的调查中国羊种布鲁氏菌rpoB基因的多态性。方法利用AMOS-PCR对分离菌株进行种型鉴定;根据布鲁氏菌rpoB基因的4个多态性位点设计特异性引物,对299株羊种布鲁氏菌流行株(羊1型菌38株、羊2型菌19株、羊3型菌242株)的rpoB基因的4个多态性位点进行扩增,并将PCR产物测序结果与羊种布鲁氏菌标准菌株的扩增结果进行比对。结果羊种布鲁氏菌的rpoB基因检测发现,有169株为rpoB-2变异型、103株为rpoB-2型、24株为新的rpoB基因型(rpoB-4型)及3株为rpoB-1型。结论我国羊种布鲁氏菌rpoB基因具有较高的多态性,可作为羊种布鲁氏菌的分子流行病学调查靶基因,我国羊种菌rpoB基因型和生物型无相关性。
ObjectiveTo investigate polymorphism of rpoB gene of Brucella melitensis strains isolated in China.Methods Species/biovars ofB.melitensisstranis were identified by AMOS-PCR. Then,specific primers were designed according to fourpolymorphic loci ofBrucella rpoBgene,four diversity loci of rpoB gene of 299 B.melitensis stranis[including B. melitensisbiovar 1(38),B. melitensisbiovar 2(19),B. melitensisbiovar 3(242)]were amplified,and sequencing results of PCRproduct were compared with rpoB gene of standard strains of B. melitensis.ResultsAnalysis of rpoB gene of 299 B. melitensisstrains revealed that 169 strains were rpoB-2 variants genotype,103 were rpoB-2 genotype,24 strains exhibited new rpoBgenotype(named asrpoB-4),3 strains of rpoB-1 genotype.Conclusion Rpo Bgene ofB. melitensisfrom China has highergenetic polymorphism,rpoBgene can be used as a target gene for molecular epidemiological investigation ofB. melitensis.
