摘要
目的:探究柚皮素对凝血功能测定过程的影响,及柚皮素在凝血诊断测试中对二价金属离子的屏蔽作用,为柚皮素在凝血诊断试剂中的应用提供理论依据。方法:分别配置不同浓度梯度的柚皮素乳浊液,加入定量标准血浆,Sysmex 1500全自动凝血分析仪分析对凝血功能检测结果的影响。每个试样测试4次,同时在添加柚皮素的前提下,在待测血浆中加入二价金属离子Cu^(2+)和Zn^(2+),测试对凝血功能检测结果的影响。结果:柚皮素的浓度低于1.5×10^(-5) mol/L时,对APTT、PT和TT无影响。柚皮素为(0.5~4.5)×10^(-4) mol/L时,检测值差异无统计学意义(P>0.05),随着柚皮素加入量的增加,APTT呈增加的趋势。在待测血浆中加入柚皮素的前提下,添加二价金属离子Cu^(2+)和Zn^(2+)与未加入上述2种离子相比,凝血测试结果差异无统计学意义(P>0.05)。结论:(0.5~4.5)×10^(-4) mol/L范围内柚皮素的加入对凝血功能检测结果差异无统计学意义,同时具有螯合二价金属离子的作用,从而屏蔽二价金属离子对凝血因子的抑制。
Objective:To investigate the significance of naringenin on blood coagulation test in order to provide theoretical basis for the application of naringenin in coagulation diagnostic reagents.Method:Low concentration and high concentration naringenin were preparedseparately with different concentration gradients,then added quantitative standard plasma and tested by Sysmex 1500 Automatic Coagulation Analyzer.Each specimen was tested 4 times.Result:It showed that there was no effect on APTT,PT and TT when the amount of naringenin was below1.5×10^-5 mol/L.When the addition of naringenin was 5×10^-5 to 4.5×10^-4 mol/L,there was no significant difference in the detection values(P〉0.05),but with the increase of naringenin added,APTT showed an increasing trend.With naringenin added to the plasma,both Cu^2+ and Zn^2+ in the plasm showed no statistical significance for coagulation tests(P〉0.05).Conclusion:The appropriate dose of naringin would have no effect on the detection result of coagulation function and havethe effect of chelating heavy metal ions,thus reducing the inhibition of Cu^2+ and Zn^2+ on coagulation factors.
