牛肝细胞再生刺激因子的制备及其生物学特性

摘要 从新生牛肝脏提取肝细胞再生刺激因子(cHSS),并对其理化及生物学特性进行研究。cHSS对原代大鼠肝细胞具有刺激活性,使DNA合成明显增加,其活性经95℃加热15分钟无明显下降,但对蛋白酶敏感。动物试验结果表明,cHSS能显著降低D-GAL致肝损伤大鼠血清ALT水平,对损伤肝细胞具有保护及修复作用,能明显提高CCL4致急性肝功能衰竭小鼠的生存率。 Hepatic stimulator substance was extracted form neonatal cattle liver and was characterized. cHSS was polypeptids with a mean molecular wight no more than 10 000. It was determined that cHSS markedly stimulated DNA synthesis of adult rat hepatocytes in primary cuture. Its stimulatory activity was not decreased obviously on heating at 95℃ for 15 min, but was lost completely on treatment with proteinase. We demonstrated that cHSS significantly lowered serum ALT leveis of D-gatactosamine-intoxicated rats and had pretctive effect on the injured liver cells. We aLso found that it markedly reduced the mortality of rats with acute hepatic failure induced by CCL4. Our findings suggest that cHSS has high stimulatory activity on regeneration of hepatocytes and may be useful clinicaly for treatment of patients with liver disease.
