摘要 脂肪酸代谢异常是恶性肿瘤细胞维持其生长需求的独特代谢方式之一,在肿瘤进展过程中发挥至关重要的作用.乙酰辅酶A羧化酶(ACC)是脂肪酸合成和氧化代谢途径的限速酶.越来越多的研究表明ACC在多种肿瘤中显著高表达,与肿瘤进展及患者预后密切相关,这使ACC可作为临床诊断及预后评估的潜在标志物;另外,抑制ACC活性可使肿瘤细胞脂肪酸合成受阻,刺激脂肪酸β氧化,使肿瘤产生严重的脂质消耗,进而抑制肿瘤生长及增殖.探讨ACC在肿瘤发生发展中的作用及分子机制,可为肿瘤靶向分子治疗带来新的思路. Abnormal fatty acid metabolism is one of the unique metabolic ways in which malignant cells maintain their growth needs and plays a crucial role in tumor progression.Acetyl-CoA carboxylase (ACC) is the rate-limiting enzyme in fatty acid synthesis and oxidative metabolism.More and more researches confirm that ACC is highly expressed in many tumors,and is closely related to tumor progression and prognosis of patients,which makes ACC as a potential marker for clinical diagnosis and prognosis.Tumor cell fatty acid synthesis can be blocked and fatty acid β oxidation can be stimulated by inhibiting the activity of ACC,resulting in serious lipid consumption of tumor,and then inhibit tumor growth and proliferation.Investigating the effect and molecular mechanisms of ACC in the genesis and development of tumors can provide a new insight into cancer targeted molecular therapy.
