生长抑制蛋白家族成员5(ING5)在人乳腺癌细胞的核质转运与不良临床病理特征的相关性分析 被引量:3

摘要 目的探究生长抑制蛋白家族成员5(ING5)在人乳腺癌细胞的核质转运及其ING5在细胞核和胞质高表达与不良临床病理特征的相关性。方法收集乳腺癌260例、淋巴结转移性乳腺肿瘤55例、乳腺纤维腺瘤61例、乳腺腺瘤病110例及癌旁乳腺组织91例。制作组织芯片,采用免疫组织化学染色法检测其ING5在细胞核和细胞质中表达情况,并分析核、质ING5蛋白与不良临床病理特征的相关性。结果乳腺癌旁组织细胞核ING5蛋白表达明显高于乳腺纤维腺瘤、乳腺腺瘤病和乳腺癌组织,腺瘤病组织细胞核ING5表达高于乳腺癌组织,乳腺癌组织细胞核ING5表达高于淋巴结转移癌细胞;癌旁乳腺组织细胞质ING5表达高于乳腺纤维腺瘤、乳腺腺瘤病和乳腺癌组织。提示ING5蛋白在人乳腺癌细胞存在核质转运。细胞核ING5高表达与远处转移呈负相关,与P53表达呈正相关,而细胞质ING5高表达与肿瘤直径和雌激素受体(ER)表达呈正相关,三阴乳腺癌(TNBC)细胞质ING5表达比非三阴乳腺癌(non-TNBC)低。结论 ING5蛋白在乳腺癌细胞存在核质转运,且细胞核ING5蛋白高表达与一些不良临床病理行为特征呈负相关。 Objective To investigate the nucleocytoplasmic translocation of inhibitor of growth family member 5 (INGS) gene in human breast cancer and the negative correlation between the nuclear high expression of ING5 protein and the clinicopathological characteristics of breast cancer. Methods We collected 260 cases of clinical primary breast cancer, 55 cases of metastatic cancer in lymph node, 61 cases of breast flbroadenoma, 110 cases of adenomatosis, and 91 cases of pare-cancerous tissues. With the tissue microarrays, we detected ING5 expression in the nucleus and cytoplasm through immunohistochemistry, and statistically analyzed the correlations of nuclear and cytoplasmic ING5 expression with the clinicopathological characteristics of breast cancer. Results The expression of nuclear ING5 in the para-csncerous tissues was obviously higher than that in the fibroadenoma, adenomatcsis and primary breast cancer; it was higher in the adenomatosis than in the primary breast cancer, and higher in the primary breast cancer than in the metastatic cancer in lymph node. The cytoplasmic ING5 expression in the para-carcinoma tissue was higher than that in the flbroadenoma, adenomatosis and primary breast cancer. These data suggested the nucleocytoplasmic translocation of ING5 protein in breast cancer. Moreover, nuclear ING5 high expression was negatively correlated with distant metastasis and positively with P53 expression, while cytoplasmic ING5 high expression was positively correlated with tumor size and estrogen receptor expression. The cytoplasm ING5 expression of triple-negative breast cancer was lower than that in the non-triple-negative breast cancer. Conclusion Nucleocytoplasmic translocation of ING5 protein occurs in breast cancer, and the high expression of nuclear ING5 is inversely related to some poor clinicopathological behaviors of breast cancer.
