同构核苷类似物的合成及其性质研究

摘要 同构核苷类似物,其光学物理性质会随着周围环境的微小变化而发生改变,被广泛应用于核酸的结构及其功能表达的研究中。运用化学合成法,设计以邻氨基苯甲酸衍生物为原料,反应得到喹唑啉衍生物碱基,此碱基与核糖反应,在醇氨溶液中水解,得到5-氯-2,4-喹唑啉二酮尿苷。并通过核磁共振波谱法、紫外可见光谱法和分子荧光光谱法确定了它的物质结构及不同极性、pH值下的紫外和荧光性质。最后通过MTT法测定5-氯-2,4-喹唑啉二酮尿苷对癌细胞增殖的影响,这为化合物在体内探针的应用提供了良好的应用前景。 Isomorphic nucleoside analogues,whose optical and physical properties change with minor changes in the surrounding environment,have been widely used in the study of nucleic acid structure and functional expression.Using the chemical synthesis method,the base of quinazoline derivative was synthesized from anthranilic acid derivatives.The base group reacted with ribose and hydrolyzed in alcohol and ammonia solution to obtain 5-chloro-2,4-quinazoline diketone uridine.The material structure and the UV and fluorescence properties at different pH values were determined by nuclear magnetic resonance spectroscopy(NMR),ultraviolet and visible spectropho tometry(UV-VIS)and molecular fluorescence spectroscopy.Finally,the effect of 5-chloro-2-azoline-4-quinazolinedione uridine on the proliferation of cancer cells was determined by MTT method,which provided a good prospect for the application of the compounds in vivo probes.
