摘要
以薏米为原料,采用大孔树脂分离纯化薏米中β-谷甾醇的粗提液,并进行抗氧化性质的研究。通过比较AB-8、D-101、DA-201三种树脂的静态吸附-解析效果,将AB-8树脂作为分离纯化的最佳树脂,并对其柱层析的吸附解析条件进行优化,进行DPPH和·OH清除能力的测定。得到β-谷甾醇分离纯化的最佳工艺条件为:树脂质量3 g,进样浓度0.55 mg/m L,75%乙醇的洗脱速度1 m L/min,此条件下得到β-谷甾醇回收率的平均值为85.56%,与预测值86.2707%接近,β-谷甾醇的纯度由原来的13.8%提高到76.17%。结果表明:AB-8树脂的分离纯化效果较好。抗氧化活性表明β-谷甾醇具有很好的抗氧化活性。
In this experiment, Coix chinensis Tod.was used as raw material, the crude extract of β-sitosterol in coix seed was separated and purified by macroporous resin, and antioxidant properties of the study. By comparing the static adsorption desorption effects of three resins,AB-8 ,D-IO1 and DA-201, Ab-8 resin as separation and purification of the best resin, the adsorption and desorption conditions of the column were optimized and scavenging ability DPPH and ·OH were determinedThe best conditions of β-sitosterol separation and purification were as follows:resin mass 3 g,injection concentration 0.55 mg/mL, elution speed 1 mL/min.Under these conditions, the average recovery of β-sitosterol was 85.56%± 0.33 % ,which was close to the predicted value of 86.2707%.The purity of β-itosterol increased from 13.8% to 76.17%.The results showed:AB-8 resin separation and purification better.Antioxidant activity indicated that/3-sitosterol had good antioxidant activity.
