摘要
目的研究永久脑缺血大鼠恢复期脑组织炎症因子动态变化规律及黄芩苷(BC)、栀子苷(GD)配伍的脑保护作用及其抗炎机制。方法 SD大鼠随机分为对照组(Con)、模型组(p MCAO)、BC∶GD(7∶3)30mg/kg、45mg/kg和60mg/kg组。线栓法建立永久中动脉闭塞模型,模拟脑缺血损伤恢复期病理过程,持续恢复3周,功能学评分观察模型动物7d、14d、21d的神经损伤情况,HE染色观察药物的脑保护作用,ELISA测定脑组织缺血半暗带区域TNF-α、IL-10、TGF-β、IL-10含量。结果黄芩苷、栀子苷灌胃给药7d-21d中均可以减少缺血区炎细胞浸润及组织水肿,降低神经功能学评分,下调TNF-α、IL-10,上调TGF-β、IL-10。结论黄芩苷与栀子苷配伍可以通过改善炎症损伤来减轻脑缺血损伤,并可调节炎性因子失衡。
Objective: To investigate the dynamic changes of inflammatory factors in the convalescent brain tissue of rats with permanent cerebral ischemia and the protective effect of baicalin(BC) and geniposide(GD) on the brain and its anti-inflammatory mechanism. Method: SD rats were randomly divided into control group(Con),model group(p MCAO),BC: GD(7: 3) 30 mg/kg,45 mg/kg and 60 mg/kg groups. The permanent middle artery occlusion model was established to simulate the pathological process of cerebral ischemia injury during the recovery period of 3 weeks. The neurological damage of 7 d,14 d and 21 d in the model animals was observed by functional score. HE staining was used to observe the protective effect of drugs. ELISA was used to determine the content of TNF-alpha,IL-10,TGF-beta and IL-10 in the ischemic penumbra region of the brain tissue. Result: BC and GD can reduce the infiltration of inflammatory cells and tissue edema in the ischemic area,reduce the score of neurologic function,down-regulate TNF-alpha,IL-10 and up regulate of TGF-beta and IL-10. Conclusion: The compatibility of BC and GD can reduce cerebral ischemic injury by improving inflammatory injury,and can modulate the imbalance of inflammatory factors.
