杂柑新品种“中柑所5号”的SSR分子标记鉴定 被引量:4

摘要 以中柑所5号及其母本爱媛30号杂柑、父本沙糖桔为材料,从22对SSR引物中筛选出适用于中柑所5号鉴定的核心引物9对,其中5对引物(CSSR038、SS16、CSSR052、SS9、CSSR014)扩增出的带型为父本型,3对引物(CSSR020、SS12、CSSR037)扩增出的带型为母本型,1对引物(SS2)扩增出异于父母本的新带型,以此建立基于SSR分子标记的中柑所5号DNA指纹图谱。利用筛选出的9对SSR引物和DNA指纹图谱,鉴定出两个疑似中柑所5号的商品苗样品确为中柑所5号。 In this study, 9 pairs of Simple Sequence Repeat (SSR) molecular marker primers were screened for construction of the fingerprint of CR1C 5 (Citrus reticulata Blanco cv. Ehime No. 30 X Citrus reticulata Blanco cv. Shiyueju). In these screened primer pairs, 5 primer pairs ( CSSR038, SS16, CSSR052, SS9, CSSR014 ) showed the same bands with its male parent Shatangju, 3 primer pairs (CSSR020, SS12, CSSR037) with its female parent Ehime No. 30, and the bond of 1 primer pair is different from both male and female parents. By using these primers for fingerprint- ing, two unknown citrus samples (from Jianyang and Ziyang, Sichuan, respectively) were identi- fied as citrus variety CRIC 5. Thus, SSR based DNA molecular marker technique is a powerful tool for discriminating this new citrus cultivar.
