摘要 目的探讨不同肝细胞肝癌病程当中miRNA602表达水平的变化,同时对二者的相关性进行分析,从分子生物学的层面为肝细胞肝癌的预防提供参考依据。方法于2013年10月至2016年6月连续收集某三级甲等医院的肝细胞肝癌确诊病例,采用Northern Blot对患者miRNA602的表达水平进行检测,并以其表达水平的中位数为临界点对所有受试者进行分组,同时对病例进行理论周期为3年的随访(若病例在随访3年时间内因肝细胞肝癌死亡则记为截尾数据)。采用Spearman等级相关及散点图分析肝细胞肝癌的不同病程与miRNA602表达水平的相关性,采用生存分析比较不同等级miRNA602表达水平的肝细胞肝癌患者的生存情况。结果本研究最终获得有效观察对象116例,肝细胞肝癌患者与非肝细胞肝癌患者的miRNA602表达水平有统计学差异(T=21.224,P<0.001),miRNA602表达水平与肝细胞肝癌病程具有统计学上的正相关性(rs=0.692,P<0.05),高miRNA602表达水平组与低miRNA602表达水平组的生存曲线差异具有统计学意义(χ2=5.725,P<0.05)。结论肝细胞肝癌的发生及病程的进展均与miRNA602表达水平有关,可考虑将miRNA602作为肝细胞肝癌的分子标志物应用于临床检测。 Objective To investigate the expression level of miRNA602 in different stages of HCC, and analyze the correlation between the two levels. Methods Hepatocellular carcinoma cases diagnosed from October 2013 to June 2016 were collected from three hospitals. The expression level of miRNA602in patientswere detected with Northern Blot. All subjects were divided into two groups according tothe median of the expression level, and performed analyses of the critical point theory within3 year follow-up. Spearman grade correlation and scatter plot were used to analyze the correlation between the different course of liver cancer and the expression level of miRNA602. Survival analysis was used to compare the survival of hepatocellular carcinoma patients with different levels of miRNA602 expression. Results 116 cases wereobtained effective observation in this study. The expression level of miRNA602 in patients with hepatocellular carcinoma was significantly different from that in non HCC patients (T = 21. 224, P 〈 0. 001), and statistically associated with hepatocellular carcinoma pathogenesis (rs= 0. 692, P 〈 0.05). Survival curves levels in high and low miRNA602 expression groupswas significantly different (χ^2 =5. 725 ,P 〈0.05). Conclusion The occurrence and progression of HCC are related to the expression level of miRNA602, miRNA602 can be used as a molecular marker for clinical detection of HCC.
