巨噬细胞极化在小鼠泡型包虫病中的作用 被引量:9

摘要 目的初步探讨巨噬细胞极化在小鼠泡型包虫病疾病进展中的作用。方法建立小鼠泡型包虫病模型。于感染后90天处死模型小鼠收集血清标本和腹腔巨噬细胞(感染组),同时收集健康小鼠的血清和腹腔巨噬细胞(对照组)。分别应用流式细胞技术和ELISA法检测M1/M2巨噬细胞及相关因子的表达情况。结果 1.流式细胞技术结果示,与正常小鼠(0.25%±0.13%)相比,感染泡球蚴90天的小鼠CD16/32^+(24.18%±3.60%)的巨噬细胞(M1)百分比呈增加趋势,P<0.05;与正常小鼠(0.30%±0.14%)相比,感染泡球蚴90天的小鼠CD206^+(5.75%±0.45%)的巨噬细胞(M2)百分比呈增加趋势,P<0.05。2.ELISA结果显示,与正常对照组相比,感染组中IL-6、TNF-α、TGF-β1的含量均有不同程度增加,P<0.05;而感染组与对照组中IL-10差异不明显,P>0.05。结论极化后的M1/M2巨噬细胞参与了泡球蚴感染过程,二者的失衡可能导致了该疾病的进展。 Objective To study the preliminarily role of maerophage polarization in the development of alveolar hydatid disease in mice.Methods The mice model of alveolar hydatid disease was established by intraperitoneal in- jection of Echinococcus multiloeularis Protoscolex.The mice were sacrificed at 90 days post-infection to collect ser- um samples and peritoneal macrophages for the infection group, while healthy mice were collected serum and perito- neal macrophages specimens for the control group.Results 1.The results of flow eytometry showed an increase in the percentage of CD16/32+ macrophages( M1 )in mice at 90 days post-infection(24.18%±3.60% )compared to normal mice (0.25%±0.13% ), P〈0.05. Moreover, The percentage of CD206+ macrophages (M2)in mice at 90 days post-in- fection(5.75%+0.45%) increased compared with the normal mice ( 0.30%±0.14%), P〈0.05.2. The results of ELISA showed that the levels of IL-6, TNF-α and TGF-β1 in the infection group increased at different degrees re- spectively compared with those in the normal control group (P〈0.05) ;The difference of IL-10 was not obvious, P〉0.05.Conclusion M1/M2 macrophages are involved in the process of Echinococcus multilocularis infection,and the imbalance between them may lead to the progression of the disease.
