摘要
探讨IL-38、IL-22和IL-17对乳腺癌发展的影响。收集34例乳腺癌患者手术前和手术后血液及27例体检健康女性血液,检测血清中的IL-38、IL-22和IL-17水平,对术前血清IL-38与IL-22和IL-17进行相关性分析并分析IL-38与乳腺癌临床病理参数之间的关系。采用不同浓度IL-38、IL-22和IL-17处理人乳腺癌MCF-7细胞,测定其细胞活力。随后分为对照组、IL-38组、IL-38+IL-22组、IL-38+IL-17组和IL-38+IL-22+IL-17组,检测其细胞活力、凋亡情况,并采用RT-PCR检测各组PCNA、Bax和Bcl-2的mRNA表达量。结果显示患者术前血清IL-38水平显著低于对照组,术后升高,与术前相比差异有统计学意义。患者术前血清IL-22和IL-17水平显著高于健康女性,术后血清IL-22和IL-17水平显著低于术前患者。患者术前血清IL-38浓度与血清IL-22和IL-17浓度均成负相关(r=-0.445,P=0.008;r=-0.498,P=0.003)。术前血清IL-38浓度与患者年龄无关,与肿瘤直径、组织学分级和淋巴结转移有关。由此表明IL-38可能与IL-17和IL-22在乳腺癌的进程中发挥相反的作用。进一步的细胞结果表明,IL-38抑制MCF-7细胞活力,促进凋亡,而IL-22和IL-17发挥相反作用。RT-PCR结果表明,IL-38促进Bax mRNA的表达,抑制PCNA和Bcl-2 mRNA的表达,而IL-22和IL-17发挥相反的作用。本研究表明IL-38抑制乳腺癌的发展,IL-22和IL-17促进乳腺癌的发展。
To explore the role of IL-38,IL-22 and IL-17 in the development of breast cancer,blood samples of 34 patients with breast cancer and 27 healthy females were collected.Blood samples were taken before and after operation.The concentration of IL-38,IL-22 and IL-17 were measured.The relationship between IL-38 and IL-22/IL-17 collected before operation were analyzed,and the relationship between IL-38 and pathological parameters were analyzed.In addition,human breast cancer cells MCF-7 were divided into 5 groups i.e.,CON,IL-38,IL-38+IL-22,IL-38+IL-17 and IL-38+IL-22+IL-17,and the cell viability was determined.Furthermore,cell viability and cell apoptosis were detected and the mRNA expression of PCNA,Bax and Bcl-2 were determined by RT-PCR.The results showed that the IL-38 concentration before operation was significantly lower than that in the healthy women,but significantly increased after operation.The levels of IL-22 and IL-17 before operation were significantly higher than those in the healthy women,nevertheless,significantly decreased after operation.Before operation serum IL-38 level was negatively correlated with IL-22 and IL-17(r=-0.445,P=0.008;r=-0.498,P=0.003).Serum levels of IL-38 was not associated with age but with tumor diameter,histological grade and lymph node metastasis.These results suggest that IL-38 may play a role in the progression of breast cancer through inhibition of IL-17 and IL-22.Further results from cancer cells treated with cytokines showed that IL-38 inhibited cell viability and promoted cell apoptosis,whereas the effect of IL-22 and IL-17 was exactly reverse of IL-38.RT-PCR results showed that IL-38 promoted the mRNA expression of Baxand inhibited the expression of PCNAand Bcl-2,while IL-22 and IL-17 showed the opposite.In conclusion,IL-38 could inhibit,while IL-22 and IL-17 could promote,breast cancer development.
