摘要
目的研究分析紫草素对甲状腺癌患者细胞转移的影响及作用机制。方法采集2016年1月—2017年1月来本院进行治疗的14例甲状腺癌患者癌组织标本进行研究。对癌细胞进行培养,MTT法分析紫草素对甲状腺癌患者癌细胞增殖的影响,流式细胞术对癌组织细胞周期分布以及凋亡情况进行分析;使用透射电镜对癌组织细胞自噬和凋亡情况进行观察。结果不同浓度的紫草素对甲状腺癌细胞均有明显的抑制作用,且抑制作用随着药物浓度的增加或者作用时间的延长而增强。不同浓度紫草素均可使甲状腺癌细胞发生明显的凋亡,且较空白对照组的细胞凋亡效果明显更好,差异有统计学意义(P<0.05),但此种凋亡作用并未随着细胞周期的进展而呈现线性变化。2μg/ml紫草素处理过24 h的甲状腺癌细胞其早期凋亡的细胞从0.2%(空白对照组)跃升至8.0%;凋亡后期继发坏死的癌细胞从0.1%(空白对照组)剧增至11.3%;紫草素处理48 h后这一现象更加明显,早期凋亡细胞为24.9%,后期继发坏死的癌细胞为14.2%。结论紫草素有明显的抑制甲状腺癌细胞增殖的作用,且与紫草素的剂量和作用时间存在明显的相关性,其主要作用机制为诱导细胞的凋亡,具有较好的抗肿瘤作用。
Objective To explore the influence of shikonin on cell metastasis in patients with thyroid carcinoma and its mechanism. Methods 14 cases of cancer tissue samples of patients with thyroid carcinoma who got treatment in our hospital during January 2016 to January 2017 were studied. The tissue were used to carry out cell cultur,MTT assay was used to analyze the influence of shikonin on reproduction of cancer cells in patients with thyroid cancer,and the flow cytometry was used to analyze the distribution of cell cycle and apoptosis of cancer tissue; and observed autophagy and apoptosis of cancer cell under electron microscopy. Results The MTT assay results showed that despite the use of different concentrations of shikonin on cancer cell function,and the effect of varying lengths of time,but we found thyroid cancer cells were significantly inhibited,and the inhibition was prolonged with the increase of drug concentration and action time. Different concentrations of porphyrin could induce apoptosis in thyroid cancer cells. The effect of apoptosis was significantly better than the blank control group(P〈0.05). However,this apoptosis did not change linearly with the progress of cell cycle. Compared with the control group,the early apoptosis of cells raised from 0. 2%(the control group) to 8% after treated by shikonin for 24 h at the dose of 2 μg/ml,and the late apoptosis cells elevated from 0.1%(control group) to 11.3%. The phenomenon of apoptosis was more obvious after been treated by shikonin for 48 h,the early apoptotic cells account for 24. 9%,and the secondary necrotic cells were 14. 2%. Conclusions Shikonin has obvious inhibitory effect on the proliferation of thyroid cancer cells,and is correlated to the dosage and time of treatment by shikonin. Its main mechanism is the induction of cell death,and has a good anti-tumor effect.
