乙酰葛根素参与大鼠脑缺血再灌注保护作用的抗凋亡介导机制研究 被引量:1

摘要 目的研究乙酰葛根素对脑缺血再灌注大鼠的保护作用及其相关机制。方法采用线栓法建立体内脑缺血再灌注模型。TTC染色缺染区面积表示梗死体积的大小,Western blot法测定beclin-1、bcl-2和bcl-xl蛋白的含量及乙酰葛根素对其影响。结果与模型组相比,乙酰葛根素可以减小梗死体积;降低beclin-1的表达,同时增加Bcl-2、Bcl-xl蛋白的表达,并呈浓度依赖性。结论乙酰葛根素通过升高抗凋亡相关蛋白发挥对脑缺血再灌注模型鼠的保护作用。 Objective To study the protection of acetylpuerarin on the middle cerebral artery occlusion(MCAO) damage and its mechanism.Methods Rats were cultured to established MCAO,2,3,5-Triphenyl-2 H-tetrazolium chloride(TTC) staining were used to calculate the volume of infarct sections,the expression of anti-apoptosis protein beclin-1,bcl-2 and bcl-xl protein were determined with Western blot and analyzed the effect of acetylpuerarin. Results Compare to model group,acetylpuerarin can attenuate the volume of infarctus,decrease the expression of beclin-1 and increase the expression of bcl-2 and bcl-xl protein. Conclusions Protective effects of acetylpuerarin on cerebral ischemia-reperfusion model rats by increasing anti-apoptosis-related proteins.
