清热利湿方及虎杖苷对SD大鼠肛旁创面肉芽组织血管通透性及Src、VE-cadherin细胞因子表达的动态影响 被引量:3

摘要 目的研究清热利湿外洗方(虎杖、蒲公英、苦参、黄柏)及虎杖苷对SD大鼠肛旁创面肉芽组织血管通透性及Src、VE-cadherin细胞因子表达的动态影响。方法以SD大鼠肛旁创面模型为研究对象,肉眼及电镜观察创面渗出、炎症及愈合情况;在术后第3、7、11天通过伊文氏蓝染色法观察创面毛细血管通透性情况;同时通过Elisa法检测创面肉芽组织Src、VE-cadherin含量。结果肉眼观察发现与生理盐水相比,清热利湿外洗方与虎杖苷均能更有效控制创面炎性渗出,促进创面愈合。造模后第3天清热利湿方组与虎杖苷组伊文氏蓝渗出情况比较差异无统计学意义(P>0.05),但二者渗出均少于生理盐水组(P<0.05);第7,11天3组伊文氏蓝渗出比较差异无统计学意义(P>0.05)。Elisa结果显示术后各阶段生理盐水组创面肉芽组织Src含量均最高(P<0.05);第3、7天清热利湿方组与虎杖苷组比较差异无统计学意义(P>0.05),术后第11天清热利湿方组Src含量高于虎杖苷组(P<0.05)。术后第3、7天各组肉芽组织中VE-cadherin浓度比较差异无统计学意义(P>0.05);术后第11天,生理盐水组VE-cadherin浓度低于其他2组(P<0.05),虎杖苷组VE-cadherin浓度低于清热利湿组(P<0.05)。结轮清热利湿方与虎杖苷可能通过抑制Src表达,调控Src/VEcadherin信号通路,进而降低血管通透性,控制创面炎性渗出,促进创面愈合。 Objective To explore dynamic effect of Chinese herbal(polygonum cuspidatum, dandelion, sophora flavescens, yellow cypress) compound and polydatin on vasopermeability and the expression of Src, VE-cadherin in SD rats models. Methods The perianal wound model of SD rats was used to observe the exudation, inflammation and healing of the wound with naked eye and electron microscope, and the capillary permeability of the wound was observed by Yi Wen's blue staining on the 3 rd, 7 th and 11 th day after operation. At the same time, the content of Src and VE-cadherin in granulation tissue was detected by Elisa method. Results There was statistical difference in vasopermeability between the 3 groups on the 3 rd postoperative day(P〈 0.05). Elisa results showed that the Src content in each stage of saline group of granulation tissue were the highest(P〈 0.05); On the 3 rd, 7 th day, TCM group and polydatin group had no significant difference(P〉 0.05), on the 11 th day, the expression of Src in TCM group was higher than that of the polydatin group(P 〈0.05). 3 groups had no significant statistically differences in the expression of VE-cadherin on the 3 rd and 7 th day(P〉 0.05); the expression of VE-cadherin in salin group was lower than the other two groups(P 〈0.05), meanwhile, polydatin group was lower than that of TCM group(P 〈0.05). Conclusion It is possible that Qingre Lishi decoction and polydatin can inhibit Src expression, regulate Src/VEcadherin signaling pathway, reduce vascular permeability, control inflammatory exudation of wound, and promote wound healing.
