摘要 黄皮白肉火龙果,又名麒麟果,为建立其快繁体系,本研究选取种子萌发长出的茎段为外植体,研究其不同消毒时间、不同浓度激素配比对其愈伤组织及芽的诱导、增殖继代、根生长的影响。结果表明:用无水乙醇灭菌40 s后,再用5%次氯酸钠溶液灭菌10 min,种子萌发率最高可达97%。愈伤组织及不定芽诱导培养基最佳配方为1/2 MS+2.0 mg/L 6-BA+1.0 mg/L NAA,芽的诱导率最高可达97%;利用MS+2.0 mg/L 6-BA+3.0 mg/L NAA培养基进行增殖、继代培养的效果最佳,增值系数最高为4.6;在MS培养基中添加2.0 mg/L IBA,可最大程度地促进根的生长。 Selenicereus megalanthus is also known as yellow pitahaya. In order to establish a system of Selenicereus megalanthus tissue culture and rapid propagation, seed germination stem segments were used as the explants, and the effects of different disinfection time, different concentrations of hormones were studied on the callus germination, propagation, and the growth of the root. The result indicated that the germination rate was 97% by sterilization with pure ethanol in 40 s, 5% hypochlorite in 10 min. The suitable medium for callus germination was 1/2 MS+2.0 mg/L 6-BA+1.0 mg/L NAA, and the germination rate was 97%; The best medium for proliferation was MS+2.0 mg/L 6-BA+3.0 mg/L NAA, and the proliferation coefficient was 4.6; The best medium for rooting was MS+2.0 mg/L IBA.
