玉屏风颗粒对地塞米松诱导的免疫抑制幼鼠的影响 被引量:10

摘要 目的探讨玉屏风颗粒对地塞米松致免疫抑制幼鼠的免疫调节作用。方法 BALB/c幼鼠(3周龄)随机分为4组,即正常对照组、模型组、玉屏风组和玉屏风+模型组,每组各10只,正常对照组和模型组均灌胃给予饮用水,持续21 d;玉屏风组和玉屏风+模型组灌胃给予玉屏风颗粒(1.8 g/kg,一日1次),持续21 d;模型组和玉屏风+模型组均分别于试验第15天和第16天腹腔注射地塞米松(12 mg/kg,一日1次)。末次给药后,小鼠称重,处死,取脾脏和胸腺称重计算脏器指数,并分离脾细胞检测自然杀伤(NK)细胞杀伤活性和CD4^+/CD8^+比值,取腹腔巨噬细胞分离纯化进行功能检测。结果与正常对照组比较,模型组幼鼠体质量、胸腺和脾脏指数、淋巴细胞增殖活性、NK细胞杀伤活性、巨噬细胞吞噬指数和脾淋巴细胞CD4^+/CD8^+比值降低,脾淋巴细胞凋亡比例升高;与模型组比较,玉屏风+模型组幼鼠体质量、胸腺和脾脏指数、淋巴细胞增殖活性、NK细胞杀伤活性、巨噬细胞吞噬指数和脾淋巴细胞CD4^+/CD8^+比值均明显升高,脾淋巴细胞凋亡比例显著降低,差异均具有统计学意义(P<0.05或P<0.01)。结论玉屏风颗粒对地塞米松诱导的幼鼠免疫抑制在不同环节均有显著的改善作用。 Objective To investigate the immunomodulatory effects of Yupingfeng granule on the immunosuppression induced by dexamethasone in immature mice. Methods BALB/c mice(3 weeks old) were randomly divided into four groups: control group, model group, Yupingfeng group and Yupingfeng +model group, ten mice in each group. The control group and model group were given by gavage once a day for 21 d with drinking water; The Yupingfeng group and Yupingfeng +model group were given by gavage once a day for 21 d with Yupingfeng granule(1.8 g/kg); the model group and Yupingfeng+model group were given by intraperitoneal injection on day 15 and 16 with dexamethasone(12 mg/kg). After last administration, the mice were weighed and sacrificed, the organ dexamethasone in spleen and thymus were measured. Meanwhile, the spleen cells and the peritoneal macrophage were isolated and purified for cell function test. Also, the nature kill(NK) cells and CD4+/CD8+ T cells were isolated from mice blood for testing. Results Compared with the control group, body weight, thymus and spleen index, lymphocyte proliferation activity, NK cell killing activity, macrophage phagocytosis index and spleen lymphocyte CD4+/CD8+ratio in model group were decreased, and the proportion of spleen lymphocyte apoptosis in model group was increased. Compared with the model group, the body weight, thymus and spleen index, lymphocyte proliferation activity, NK cell killing activity, macrophage phagocytosis index and spleen lymphocyte CD4+/CD8+ ratio in Yupingfeng+model group were increased, and the proportion of spleen lymphocyte apoptosis in Yupingfeng+model group was decreased, the differences were statistically significant(P〈0.05 or P〈0.01). Conclusion Yupingfeng granule can significantly improve the immune function of the immunosuppressive immature mice induced by dexamethasone.
