摘要 目的探讨表皮生长因子样结构域(EGFL7)和黏着斑激酶(FAK)在婴幼儿血管瘤组织中的表达及相关性。方法采用免疫组织化学(SP法)方法检测婴幼儿血管瘤和正常皮肤组织中EGFL7和FAK的表达水平;培养脐静脉内皮细胞(HUVEC),用Western-blot方法,检测在不同重组人EGFL7浓度下,EGFL7和FAK蛋白的表达情况。结果婴幼儿血管瘤增生期组织中EGFL7和FAK的表达明显高于消退期和正常皮肤组织(P<0.05),而消退期组和正常皮肤组比较差异无统计学意义(P>0.05),EGFL7和FAK蛋白的表达与脐静脉内皮细胞(HUVEC)中重组EGFL7的浓度呈正相关。结论 EGFL7和FAK可能通过影响血管形成参与了婴幼儿血管瘤的发展,EGFL7蛋白可能成为预测婴幼儿血管瘤的病情发展及指导临床治疗的标志物之一。 Objective To discuss the expression of EGFL7 and FAK and also analyse their correlation in infantile hemangioma. Methods The expression level of EGFL7 and FAK in both infantile hemangioma and normal skin tissues were detected by Immunohistochemistry(SP method). The expression of EGFL7 and FAK in cultured umbilical vein endothelial cells was detected by Western-blot method at different concentrations of recombinant human EGFL7. Results The expression of EGFL7 and FAK in the proliferative phase of infants with hemangioma was significantly higher than that in regression group and normal skin tissues(P〈0.05). There was no significant difference between the regression group and the normal skin group(P〉 0.05). The protein expression level of EGFL7 and FAK were positively correlated with the concentration of EGFL7 in umbilical vein endothelial cells(HUVEC). Conclusion EGFL7 and FAK may be involved in the development of infantile hemangioma by influencing angiogenesis, and EGFL7 protein may be one of the markers for predicting the development of infantile hemangioma and guiding clinical treatment.
