抗体应答研究技术在非人灵长类感染与免疫模型中的应用

Application of Antibody Response Research Technologies in Non-human Primate Infection and Immunization Models

摘要 近年来,随着单个B细胞抗体筛选技术与深度测序(Deep Sequencing)的发展,相应技术被广泛应用于人体内抗体应答研究中。但该类技术在非人灵长类(Non-Human Primates,NHPs)抗体应答研究中的应用较为有限。而NHPs由于与人类具有较为相似的免疫系统,二者在感染或免疫后可以激起相似的获得性免疫应答,尤其是对NHPs产生的针对病原体感染或疫苗的NHP抗体应答谱研究,可以为人类免疫学研究、疫苗设计及生物药物临床转化提供关键信息。本文对目前常用非人灵长类抗体筛选技术的研究进展与应用实例进行了综述,以期为生物大分子药物研究者提供较为完备的参考。 Development of technologies based on cloning of single B-cell antibodies as well as deep sequencing has led to their wide use in the study of human antibodies.However,these approaches are limited in antibody-response research in non-human primates(NHPs).Due to the relative similarity in immune systems,a similar adaptive response can be elicited after infection/immunization in humans and NHPs.Therefore,an investigation on the antibody profile in a humoral immune response stimulated by viral infection or vaccination should provide important information for human immunological studies,vaccine design,and the translation from basic studies to clinical practice.In this review,we describe technologies for the study of NHP monoclonal antibodies and their application.
