摘要 目的通过饲喂高脂饲料建立高血脂小鼠模型,并给予血清鸢尾素(Irisin)处理,探索Irisin对小鼠血脂水平的影响,为Irisin的临床应用提供依据。方法 24只8周龄雄性昆明小鼠,采用完全随机设计,根据随机数表法分为三组,每组8只:普通膳食对照组[普通饲料+磷酸盐缓冲液(PBS)]、高脂膳食模型组(高脂饲料+PBS)和Irisin干预组(高脂饲料+0.125 ng/kg Irisin,用等量PBS溶解)。饲养2周,记录体质量,同时Irisin干预组每周给予2次尾静脉注射Irisin处理,最后一次处理后隔夜取血,用甘油三酯(TG)试剂盒、总胆固醇(T-CHO)试剂盒和高密度脂蛋白(HDL-C)试剂盒检测小鼠的血脂变化水平。结果实验结束后,高脂膳食模型组中小鼠体质量达(42.28±1.27)g、血清TG含量为(0.86±0.21)mmol/L、T-CHO含量为(6.79±0.86)mmol/L,HDL-C含量为(6.18±0.85)mmol/L,分别与普通膳食对照组的(40.65±1.26)g、(0.69±0.10)mmol/L、(3.25±0.54)mmol/L和(8.49±1.69)mmol/L比较差异均有统计学意义(P<0.05);Irisin干预组小鼠的体质量为(40.54±1.34)g,而血清中TG和HDL-C含量分别为(0.59±0.10)mmol/L和(8.03±1.54)mmol/L,与高脂膳食模型组相比差异均有统计学意义(P<0.05)。结论血清Irisin对小鼠血脂代谢水平有显著改善。 Objective To provide the basis for clinical application of irisin through investigating the effects of irisin on lipid metabolism of mice by establishing hyperlipidemia models induced by high-fat diet and disposed of serum irisin. Methods A total of 24 eight-week male Kunming mice were divided into 3 groups according to random number table method as follow: the diet control group(normal diet + phospate butfer [PBS]), the high fat diet model group(highfat diet + PBS), the irisin intervention group(high-fat diet + 0.125 ng/kg irisin dissolved by PBS), with 8 mice in each group. During 2 weeks of feeding, the irisin intervention group was performed by the intravenous injection with corresponding dose of irisin through vena caudalis twice a week and the weight was recorded. The blood samples were taken after overnight after the last injection, the changes in lipid metabolism were tested with triglyceride(TG), cholesterol total(TCHO) and high-density lipoprotein(HDL-C) kits. Results There were significant difference between the diet control group(weight:(40.65±1.26) g; TG :(0.69±0.10) mmol/L; T-CHO:(3.25±0.54) mmol/L; HDL-C:(8.49±1.69) mmol/L)and the high-fat diet model group(weight:(42.28±1.27) g; TG(0.86±0.21) mmol/L; T-CHO:(6.79±0.86) mmol/L; HDLC:(6.18±0.85) mmol/L) in difference indexes(P〈0.05). It meant that the hyperlipidemia model was successfully established. There also were significant differences between the high-fat diet model group and the irisin intervention group(weight:(40.54±1.34) g; TG(0.59±0.10) mmol/L; HDL-C:(8.03±1.54) mmol/L) in some indexes(P〈0.05). Conclusion The effects of serum irisin on the level of lipid metabolism improvement in mice are significant.
