桑葚花色苷提取物对高脂酒精膳食作用下小鼠棕色脂肪组织改变的影响 被引量:5

Effects of cyanidin-3-O-β-glucoside extracted from mulberry on brown adipose tissue of mice fed with high fat and alcohol diet

摘要 目的研究桑葚花色苷提取物对高脂酒精膳食作用下小鼠棕色脂肪组织改变的影响。方法将体重23 g左右的C57BL/6J雄性小鼠随机分为四组:正常对照组、花色苷提取物组、高脂酒精模型组和高脂酒精+花色苷提取物组,前两组喂食普通饲料,后两组喂食高脂饲料,酒精添加到饮水中,花色苷以灌胃的方式给予。喂养12周后处死小鼠,收集脂肪组织样本,称重,做HE染色以及荧光定量PCR分析。结果桑葚花色苷提取物对高脂酒精膳食作用下小鼠棕色脂肪组织的脂肪系数无影响,但可以降低高脂酒精膳食作用下小鼠棕色脂肪组织中小直径脂滴空泡的比例,由(74.12±1.90)%降低至(60.63±2.21)%,并升高高脂酒精膳食作用下棕色脂肪组织中等直径脂滴空泡的比例,由(24.72±1.42)%升高至(31.65±1.43)%,差异均有统计学意义(P<0.05)。同时提高其功能性基因解偶联蛋白1的表达,差异有统计学意义(P<0.05)。结论桑葚花色苷提取物可以改善由高脂酒精膳食所引起的小鼠棕色脂肪组织的形态学改变和功能抑制,为花色苷防治慢性代谢性疾病提供新思路。 Objective To study the effects of cyanidin-3-O-β-glucoside extracted from mulberry on brown adipose tissue of mice fed with high fat and alcohol diet. Methods The mice were randomly divided into 4 groups which were control group,cyanidin-3-O-β-glucoside group,high fat with alcohol group and high fat with alcohol plus cyanidin-3-O-β-glucoside group.The first two groups were fed with chow diet,the last were fed with high fat diet receiving alcohol in drinking water and orally gavaged with cyanidin-3-O-β-glucoside solution per day for 12 weeks. Then mice were sacrificed and tissue samples were collected for HE staining and fluorescent quantitative real-time PCR analysis. Results Cyanidin-3-O-β-glucoside has no effect on aliphatic index of brown adipose tissue but could significantly improve the distribution of lipid droplet size compared to the high fat with alcohol group from(74.12±1.90)% to(60.63±2.21)% in ratio of small diameter lipid droplets(P〈0.05)and from(24.72 ± 1.42)% to(31.65 ± 1.43)% in ratio of middle diameter lipid droplets(P〈0.05). Moreover cyanidin-3-O-β-glucoside could markedly increase the expression of UCP1 repressed by high fat with alcohol diet(P〈0.05).Conclusion Cyanidin-3-O-β-glucoside extracted from mulberry could significantly improve the histological structure and the expression of thermogenic gene in brown adipose tissue of mice which was effected by high fat with alcohol diet. This study provides a new idea for preventing chronic metabolic diseases.
