摘要
目的探讨白藜芦醇(Res)对卵巢癌细胞株SKOV-3增殖、迁移和侵袭的影响及可能机制。方法培养SKOV-3细胞,将细胞分为对照组(Control)与白藜芦醇组(Res),采用四甲基偶氮唑蓝(MTT)法检测白藜芦醇对SKOV-3细胞活力的影响;应用Transwell小室法检测SKOV-3细胞迁移和侵袭能力的变化;应用Real-time PCR方法检测两组细胞内Akt mRNA、Bax mRNA与bcl-2 mRNA的表达水平;应用Western blot方法检测两组细胞内Akt、Bax与bcl-2蛋白表达。结果 MTT检测与Transwell实验结果发现,白藜芦醇能够显著抑制SKOV-3细胞的增殖、迁移和侵袭能力。Real-time PCR方法与Western blot方法检测发现,白藜芦醇能够显著上调Bax mRNA与蛋白表达水平,明显抑制Akt与bcl-2 mRNA及蛋白的表达水平。结论白藜芦醇抑制卵巢癌细胞株SKOV-3增殖、迁移和侵袭可能与上调Bax,抑制Akt与bcl-2蛋白表达相关。
Objective To investigate the effect and possible mechanism of Resveratrol(Res) on the proliferation, migration and invasion of ovarian cancer cells line SKOV-3. Methods SKOV-3 cells were divided into control group(Control) and Resveratrol group(Res). MTT method was used to detect the effects of Resveratrol on SKOV-3 cell viability; migration and invasion of SKOV-3 cells were observed by Transwell assay; the expression of Akt mRNA, Bax mRNA and bcl-2 mRNA in SKOV-3 cells was detected by real-time PCR; the expressions of Akt, Bax and Bcl-2 proteins in SKOV-3 cells were detected by Western blot. Results The proliferation, migration and invasion of SKOV-3 cells were inhibited significantly after treatment with Resveratrol, and Resveratrol significantly up-regulated the expression level of Bax mRNA and protein, and significantly inhibited the expression level of Akt and Bcl-2 mRNA and protein. Conclusion Resveratrol could inhibit proliferation, migration and invasion of ovarian cancer cell line SKOV-3, which might be related to up-regulation of Bax and inhibition of expression of Akt and bcl-2 proteins.
